摘  要:目的研究UVB辐射对黑色素细胞增殖、黑素含量、酪氨酸酶活性及黑素合成相关基因表达的影响。方法原代分离培养黑色素细胞,采用MTT法测定不同强度UVB干预后细胞增殖率,L-DOPA法检测细胞中酪氨酸酶活性,NaOH裂解法测定黑素含量,RT-PCR检测黑素合成相关基因MITF、TRY、TRP-1、TRP-2的表达,以未经UVB辐照的细胞为空白对照组。结果与空白对照组相比,0.3J/cm^2 UVB模型组细胞增殖率显著增高(P<0.01),酪氨酸酶活性明显提高,差异有统计学意义(P<0.05),细胞中黑素含量有显著增加的趋势,差异有统计学意义(P<0.01)。0.3J/cm2 UVB干预后能显著上调细胞中黑素合成相关基因MITF、TRY、TRP-1、TRP-2mRNA的表达。结论 UVB辐射对黑素合成途径中相关基因的表达具有正向调控作用,细胞中黑素含量明显增加,对构建和优化理想的色素沉着细胞模型提供了参考依据。Objective To study the UVB radiation on melanoma cell line proliferation,melanin content and tyrosinase activity and related gene expression.Methods After different intensities of UVB intervention to primary cultured melanocytes,the cell proliferation rate was detected by MTT assay.Cells tyrosinase activity was detected by L-DOPA assay and melanin content was detected by NaOH lysis assay.RT-PCR was used to detectthe expression of mRNA of MITF,TRY,TRP-1,TRP-2.Without UVB irradiation group was the control group.Results Compared to the control group,in 0.3J/cm2 UVB intervention melanocytes group,cell proliferation rate and tyrosinase activity increased significantly,and the difference were statistics significance(P〈0.01 and P〈0.05).And cell melanin content had significantly increasing trend,and the difference was statistically significant(P〈0.01).0.3J/cm2 UVB intervention can significantly increase the mRNA expression of MITF,TRY,TRP-1,TRP-2in cells.Conclusion UVB radiation has a positive effect on the regulating genes expression which are related to the melanin synthesis pathway and the melanin significantly increased in the cells,which provide more information for constructing pigmentation models.
