抑制PI3K通路增强CAHB对Jurkat细胞的凋亡作用

摘　　要：目的探讨CAHB单独使用及与LY294002联合应用对Jurkat细胞体外作用的影响。方法分别用不同剂量的CAHB(0、5、10、20、40mmol/L)诱导Jurkat细胞,用MTS法检测Jurkat细胞的增殖抑制效应,流式细胞术分析Jurkat细胞凋亡情况;取40mmol/L浓度的CAHB加入PI_3K抑制剂LY294002,流式细胞术分析Jurkat细胞凋亡情况。结果 CAHB对Jurkat细胞生长有显著的抑制作用,并能显著诱导Jurkat细胞发生凋亡,其作用呈明显剂量、时间依赖性。PI_3K抑制剂LY294002能显著增强CAHB对Jurkat细胞的凋亡作用。结论 CAHB能抑制Jurkat细胞增殖,并诱导其凋亡。抑制PI_3K通路能增强CAHB对Jurkat细胞的凋亡作用。Objective To study CAHB alone or in combined with LY294002 on culture Jurkat cell lines and the possible mechanism. Methods Jurkat cells were treated with different concentrations of CAHB in culture. MTS assay was used to observe the proliferated inhibition rate of Jurkat, and the flow cytometry was applied to analyze rate of AnnexinⅤ＋/PI-, and to observe the change in the rate of AnnexinⅤ＋/PI- in presence or absence of PI3K inhibitor LY294002 after Jurkat cells were induced with CAHB. Results CAHB obviously inhibited the growth of Jurkat cells in a time-and dose-dependent manners. CAHB also induced apoptosis of Jurkat cells in a dose-dependent manner. The PI3K inhibitor LY294402 markedly increased the rate of AnnexinⅤ＋/PI- of Jurkat cells which was induced with CAHB. Conclusion CAHB can inhibit the proliferation and induce apoptosis of Jurkat. Inhibition of PI3K signal enhances sensitivity of Jurkat cells to CAHB.
