摘 要:目的探讨PRP联合h UC-MSCs移植对大鼠骨质疏松性骨折愈合的影响。方法雌性SPF级SD大鼠50只随机分为5组,每组10只:假手术组(仅切除卵巢周围组织),安慰剂组(注射生理盐水),未诱导组(给予未诱导的h UC-MSCs),成骨诱导组(移植诱导后的h UC-MSCs),PRP+成骨诱导组(移植诱导后的h UC-MSCs联合PRP)。OPF造模成功后,对各组动物进行细胞移植3次,分别在OPF造模后1周、3周和7周。骨折后的第1周、3周、5周、7周、11周、13周Micro CT扫描动物模型患肢。结果 Micro CT动态观察结果显示,PRP+成骨诱导组骨痂生成量明显多于、早于其余各组,骨折11周时达到临床愈合标准,而其他组别骨痂重塑期相对滞后,比正常骨量大鼠愈合时间至少推迟了37.5%。结论 PRP联合h UC-MSCs移植可促进大鼠骨质疏松性骨折愈合,为临床治疗OPF提供数据支持。Objective To investigate the effect of PRP combined with h UC-MSCs transplantation on the healing of osteoporotic fracture in rats.Me thods Fifty female SPF grade SD rats were randomly divided into 5 groups,10 rats in each group:The sham operation group was only resected ovarian tissues;The placebo group was injected with saline;The non-induction group transplanted withuninducedh UC-MSCs;The PRP + induction group transplanted withh UC-MSCs combined with PRP.After the OPF model was successfully constructed,cells were transplanted 3 times in each group,1 week,3 weeks and 7 weeks after fracture.Micro-CT analysis was performed every two weeks after fracture.Re sults Micro-CT analysis results showed that the amount of callus formation in PRP + induction group was significantly increased than that in the other groups.Clinicalhealing criteria was achieved at11 weeks post-fracture,while other groups callus remodeling phase is lagging behind.The bone healing time in rats was delayed for at least 37.5% compared with normal fracture animals.Conclusion PRP combined with h UC-MSCs transplantation promote osteoporotic fracture healing in rats.
