摘 要:目的 探讨模拟失重30 d后高+Gx暴露对猴牙龈组织结构和热休克蛋白70 (heatshock protein 70,HSP70)表达的影响. 方法 11只雄性猕猴按随机数字表法分为4组,即正常对照组(A组,2只)、30 d模拟失重组(B组,3只),+13 Gx暴露组(C组,3只)、模拟失重30 d后+Gx暴露组(D组,3只).其中D组根据+Gx峰值又分为+11 Gx/270 s(D1)、+13 Gx/230 s(D2)和+15 Gx/200 s(D3)3组,每组1只.动物放血处死,组织取材后,经4%甲醛固定,石蜡包埋切片,采用组织病理学和免疫组织化学的方法,观察猴牙龈组织结构及HSP70的表达情况. 结果 A组可见正常的牙龈组织结构,其它组牙龈组织结构基本正常,无明显改变.A组牙龈组织HSP70呈阴性表达,上皮和结缔组织均无明显着色;B、C组呈弱阳性表达,牙龈上皮着色弱阳性,结缔组织无明显着色;D组牙龈组织呈阳性表达,牙龈上皮着色阳性,结缔组织无明显着色.统计学显示D组比B组和C组阳性表达量明显增高(P<0.01),D3组比D1和D2组阳性表达量明显增高(P<0.01). 结论 30 d模拟失重和高+Gx暴露都诱导猴牙龈组织产生HSP70,模拟失重后高+Gx暴露显著增加了HSP70的表达.Objective To study possible influence of high +-Gx exposure on HSP70 expression in gingiva of the rhesus monkey after 30-day simulated weightlessness.Methods Eleven male rhesus monkeys were randomly divided into four groups:control groups (A,n=2),30 d simulated weightlessness group (B,n=3),+13 Gx group (C,n=3) and high +Gx exposure after 30 d simulated weightlessness group (D,n=3).The group D was divided into 3 subgroups according to Gx peak loads:+11 Gx/270 s (D1),+13 Gx/230 s (D2) and +15 Gx/200 s (D3).The tissues of sacrificed animals were sampled and were fixed with 10% buffered formaldehyde and embedded in paraffin.The paraffin-embedded sections were analyzed by histopathological and immunohistochemical methods to observe the gingival tissue structure and the expression of HSP70.Results Group A showed normal structure of the gingival tissue and other groups' were also normal and without obvious change.Group A showed negative expression of HSP70 that the gingival epithelium and connective tissue had no significant stain detected;Group B and C expressed weak positive expression that the stain of gingival epithelium was weak positive and the connective tissue had no significant stain.The gingival tissues of group D showed positive expression that the stain of gingival epithelium was positive and the connective tissue had no significant stain.The expression quantity of HSP70 in the D group was significantly higher than that in group B and C (P〈0.01).Group D3 had significantly higher positive expression quantity than group D1 and D2 (P〈0.01).Conclusions Thirty-day simulated weightlessness and high +Gx exposure would induce the significance of HSP-70 expression in gingival tissues of rhesus monkey.
