摘 要:目的建立失血性休克大鼠动物模型,观察紫芪方对失血性休克大鼠肠上皮细胞线粒体损伤的保护作用。方法将大鼠随机分为休克组、复苏组、紫芪方组、参附组、蒸馏水组,每组6只;采用改良Wigger's方法制备大鼠失血性休克模型,观察紫芪方对模型大鼠肠上皮细胞线粒体呼吸功能、膜电位、Na^+-K^+-ATP酶活性、细胞色素氧化酶活性的影响。结果与休克组及单纯复苏组比较,紫芪方组和参附组肠上皮细胞线粒体呼吸功能增强,线粒体Na^+-K^+-ATP酶活性显著升高,肠上皮细胞罗丹明123负载实验荧光强度显著增强,而且紫芪方组可使线粒体呼吸功能、Na^+-K^+-ATP酶活性和细胞色素氧化酶活性恢复至正常水平更明显。结论紫芪方可改善大鼠肠上皮细胞线粒体功能,稳定膜电位,提高Na^+-K^+-ATP酶及细胞色素氧化酶活性,对失血性休克肠上皮损伤具有保护作用。Objective To establish a model of rats with hemorrhagic shock in order to observe the protective effects of Ziqi compound on the intestinal epithelial mitochondria with hemorrhagic shock. Methods The rats were randomly divided into shock group,recovery group,Ziqi group,Shenfu group and distilled water group. Each group had 6rats. The hemorrhagic shock model was established using improved Wigger's method. The effects of Ziqi compound on the respiratory function,membrane potentials,Na^+-K^+-ATP enzymic activity and the cytochrome oxidase activity of the rat model were observed. Results In comparison with the shock group and the anabiosis group,the respiratory function and Na^+-K^+-ATP enzymic activity of the intestinal epithelial mitochondria and the fluorescence intensity of rhodamine123 load experiment of Ziqi compound group and Shenfu group were significantly reinforced. Moreover,the effect was more remarkable in Ziqi compound group,in which the mitochondria respiratory function,Na^+-K^+-ATP enzymic and the cytochrome oxidase activity returned to normal. Conclusion Ziqi compound can improve intestinal epithelial mitochondrial function,stabilize membrane potential,and raise the Na^+-K^+-ATP enzymic activity and the cytochrome oxidase activity,so it has significant protective effects on intestinal epithelium of hemorrhagic shock.
