摘 要:优化甘草废渣中总黄酮的提取工艺。以甘草苷为对照品采用比色法测定总黄酮含量,在单因素基础上采用Box-Benhnken响应面法以总黄酮提取得率(Y,%)为响应值,以乙醇浓度(X_1,%)提取时间(X_2,min)、液料比(X_3,mL/g)及提取次数(X_4,次)为自变量,研究各自变量及各自变量间交互作用对总黄酮提取得率的影响,对3批甘草废渣进行验证试验。研究结果表明甘草黄酮最佳提取工艺为:乙醇浓度为70%、提取时间120 min、液料比10:1(mL/g)、提取次数2次,在优化条件下对3批甘草废渣进行工艺验证,甘草黄酮提取得率为(2.4±0.34)%(n=3)与预测理论值2.39%接近。利用Box-Behnken响应面法优化得到的甘草废渣总黄酮提取工艺参数可靠,方法简便,预测性良好。Extraction parameters of total flavonoids from licorice residue was optimized by response surface method. The total flavonoids content was determined by colorimetric method. The effect ofethanol concentration,extraction time, the ratio of material to liquid ratio and extraction times was studied. The effect of each factor and interaction on the extraction rate of total flavonoids was studied. 3 batches of licorice residue were used for validation test under the optimized conditions. Research results showed that thebest extraction parameters was:ethanol concentration 70 %, extracting time 120 min, ratio of 10 ∶ 1(m L/g), extracting times of 2, the extraction rate of flavonoids was(2.4±0.34) %(n=3), the difference was little between 2.39 % predictions.The extraction parameters of total flavonoids from licorice residue obtained by Box-Behnken response surface method were reliable, simple and convenient.
