摘 要:建立了一种高效液相色谱法(HPLC)测定角鲨烯含量的检测方法。采用XBridge RP C18色谱柱(250 mm×4.6 mm,3.5μm),流动相为甲醇,流速为1.0 m L/min,检测器为光电二极管阵列检测器,检测波长为210 nm。结果表明,在上述色谱条件下,角鲨烯浓度在2.6μg/m L^260μg/m L范围内线性关系良好,相关系数r为0.999 9。检出限(S/N=3)为0.33μg/m L,定量限(S/N=10)为0.99μg/m L,精密度RSD(n=6)为0.40%;平均加标回收率为100.78%,相对标准偏差为0.75%。方法准确、灵敏、可靠,可用于角鲨烯的定量和定性分析。A high-performance liquid chromatography method for the determination of squalene was developed.The determination of squalene was performed on a XBridge RP C18column(250 mm× 4.6 mm,3.5 μm). The mobile phase was methanol,and the flow rate was 1.0 m L/min.The detector was photodiode array detector and the variable wavelength detector(VWD) was set at 210 nm.In the above chromatographic conditions,good linear was observed in the linear range of 2.6 μg/m L-260 μg/m L, and the correlation coefficient r was 0.999 9.The limit of detection(LOD) of squalene was 0.33 μg/m L(S/N=3).The quantitation of detection(QOD) of squalene was 0.99 μg/m L(S/N=10).The relative standard deviation(RSD) for the accuracy was 0.40 %(n=6)and the average recovery was 100.78 % with RSD of 0.75 %.The method is sensitive,reliable,accurate and could be used for the determination of squalene.
