摘 要:目的探究骨髓间充质干细胞对大鼠心脏移植的免疫调节作用。方法选取实验动物中心健康雄性SD大鼠与Wistar大鼠,将其分成A、B、C三组,建立大鼠心脏移植模型,贴壁法分离培养大鼠MSC(骨髓间充质干细胞),由受体颈外静脉注入,观察移植后大鼠的存活时间,HE(苏木精-伊红)染色观察移植心脏淋巴细胞浸润程度。结果 B、C两组与A组对比,大鼠平均存活时间差异有统计学意义(P<0.05),B、C两组对比平均存活时间差异无统计学意义(P>0.05)。B、C两组与A组移植心脏病理学分级评分对比差异有统计学意义(P<0.05)。B、C两组移植心脏病理学分级评分差异无统计学意义(P>0.05)。结论在体外混合淋巴细胞反应中MSC能抑制T细胞增殖,但在体内实验中没有观察到移植物存活时间显著延长及没有减轻淋巴细胞浸润。Objective To explore the effect of bone marrow mesenchymal stem cells on heart transplantation in rats. Methods From the experimental animal center of healthy male SD rats and Wistar rats, divided into A, B, C three groups,to establish the model of rat heart transplantation, adherent cultured rat MSC(mesenchymal stem cells), injected by the external jugular vein of the recipient after transplantation, observe the survival time of rats, HE(hematoxylin and eosin) staining of cardiac allograft lymphocyte infiltration. Results B, C two groups compared with the A group, the average survival time difference was statistically significant(P〈0.05), B, C two groups compared with the average survival time difference was not statistically significant(P〈0.05). There was a significant difference in B, C two group and A group(P〈0.05). There was no significant difference in B and C two groups(P〉0.05). Conclusion MSC can inhibit the proliferation of T cells in vitro, but not significantly prolonged the survival time of grafts in vivo, and no significant decrease of lymphocyte infiltration.
