摘 要:目的制备芦丁磁性脂质体,并对其包封率、粒径大小和稳定性进行检测。方法采用超声薄膜分散法制备芦丁脂质体;采用化学沉淀法制备Fe3O4颗粒,并利用水合油酸钠包覆沉淀出的磁性颗粒;用超声将芦丁脂质体和磁流体混合分散,制备芦丁磁性脂质体;利用紫外可见分光光度法测定其包封率,并初步考察磁性脂质体的稳定性。结果制备的芦丁磁性脂质体在显微镜下为圆形,直径分布为29.4~39.2μm,包封率为(20.69±4.56)%,粒径大小较为集中,短时间内稳定性较好。结论用超声薄膜分散法制备的芦丁磁性脂质体平均粒径较均匀,外观呈圆形且表面光滑,分布均匀,稳定性较好,但包封率有待提高,制备后4℃保存,渗漏率较低。Objective To prepare magnetic rutin liposomes and detect its encapsulated ratio,particle size and stability. Methods The rutin liposome was prepared by ultrasound film dispersion method. The Fe3O4 magnetic fluid was prepared by the chemical coprecipitation,and coated Fe3O4 particle with acid. Lastly,mix and disperse the rutin liposome and magnetic fluid by means of ultrasonic to prepare magnetic rutin liposomes. There encapsulation efficiency was detected by the ultraviolet- visible absorption spectrometry method,and the stability of magnetic liposome was investigated. Results The magnetic rutin liposomes by orthogonal design were globular,the diameter spread is 29. 4 ~ 39. 2 μm. Its encapsulation efficiency are( 20. 69± 4. 56) %,and the particle size is more concentrated and the short- time stability is good. Conclusion The magnetic rutin liposomes have well- proportioned diameter by ultrasound film dispersion method,its appearance is globular and surface is smooth,its distribution is uniform and the stability is good. However,the encapsulation efficiency needs to be improved,and when saved at 4℃ after preparation,the leakage ratio is low in the short time.
