摘  要:目的:建立高效液相色谱(HPLC)法同时测定承德地区山里红叶中绿原酸和牡荆素鼠李糖苷的含量。方法:采用Agi Lent ZORBAX SB-C18色谱柱(4.6 mm×250 mm,5μm);以乙腈-0.1%甲酸水溶液-四氢呋喃为流动相梯度洗脱;检测波长为340 nm;检测时间50 min;流速1.0 m L·min-1;柱温30℃;进样量10μL。结果:15批承德山里红叶样品中绿原酸和牡荆素鼠李糖苷的线性范围分别为2.56-410、2.51-400 mg·L-1(r分别为0.999 9,0.999 8);平均加样回收率分别为99.8%(RSD 0.48%),99.7%(RSD 0.77%)。结论:该方法准确可靠,重现性好,可用于山里红叶的质量控制。Objective: To develop HPLC method for simultaneous determination of chlorogenic acid and vitexin rhamnoside in Major Hawthorn Leaf of Chengde. Method: With an Agilent ZORBAX SB-C18column( 4. 6 × 250 mm,5 μm),gradient elution was performed by mobile phase of 0. 1% formic acid-acetonitrile-tetrahydrofuran. The detection wavelength was 340 nm; detection time was 50 min; f Low rate was 1. 0 m L·min- 1; the column temperature was 30℃;injection volume was 10 μL. Results: In the 15 batches of Major Hawthorn Leaf of chengde,the Linear ranges of chlorogenic acid and vitexin rhamnoside were from 2. 56 - 410 mg·L- 1( r = 0. 999 9) to 2. 51 - 400 mg·L- 1( r =0. 999 8),respectively. The average recovery rates of chlorogenic acid and vitexin rhamnoside were 99. 8%( RSD0. 48%),99. 7%( RSD 0. 77%),respectively. Conclusion: This method is accurate,reproducible and suitable for controlling the quality of Major Hawthorn Leaf.
