摘  要:目的比较二(喹唑啉-4-基)二硒醚(LG003)与表柔比星、尼莫司汀、长春地辛三种抗肿瘤药物对肺癌A549细胞的体外抗肿瘤活性。方法利用MTT比色法检测LG003与三种药物对靶细胞增殖的影响;利用形态学观察、甲瓒结晶生成和乳酸脱氢酶活力测定评价其对靶细胞的细胞毒性。结果在实验浓度下,LG003对A549细胞显示出较好的抑制活性。在1μmol/L浓度下LG003对靶细胞的抑制活性低于长春地辛和表柔比星;在10μmol/L浓度下其抑制活性高于表柔比星和尼莫司汀。LG003在1~40μmol/L浓度下细胞毒性显著低于长春地辛(P〈0.05);5~40μmol/L浓度下其细胞毒性显著低于表柔比星(P〈0.05)。结论 LG003对A549细胞具有较高的体外抑制活性和较低的细胞毒性,是开发抗肿瘤药物的理想候选化合物。Objective To Compared the antitumor activities of 1,2- di( quinazolin- 4- yl) diselane( LG003) with epirubicin,nimustine and vindesine on lung cancer A549 cells in vitro. Methods Cell proliferation was measured by MTT assay. The cytotoxicity was compared by morphologic observation,formazan crystal and LDH release assay. Results Under the experiment concentration,LG003 showed better inhibitory activity of A549 cells. At the dose of 1μmol / L,the cell growth inhibitory rate of LG003 was less than that of vindesine and epirubicin. At the dose of 10μmol / L,the cell growth inhibitory rate was higher than that of epirubicin and nimustine. From 1 to 40μmol / L,the cytotoxicity of LG003 was significantly lower than that of vindesine( P〈0. 05). From 5 to 40μmol / L,its cytotoxicity was significantly lower than that of epirubicin( P〈0. 05). Conclusions The compound LG003 possesses high growth inhibitory activity but with low toxicity on A549 cells in vitro. It is a potential anticancer agent.
