摘  要:目的:研究燃煤型氟中毒大鼠睾丸雄激素结合蛋白(ABP)和转铁蛋白(Tf)表达的改变。方法:105只雄性SD大鼠为研究对象,随机分为对照组、低氟组、中氟组、高氟组、低氟加营养组、中氟加营养组及高氟加营养组,各染毒组喂饲含不同比例的地氟病区煤烘玉米的饲料(加营养组添加豆粉),构建燃煤型氟中毒动物模型;各组动物于喂养90、120及180 d分3批处死,观察氟斑牙、尿氟,免疫组织化学法观察睾丸ABP和Tf表达的变化。结果:大鼠氟斑牙分度和尿氟含量随时间及染毒剂量增加逐渐加重(高氟组>高氟加营养组>中氟组>中氟加营养组>低氟组>低氟加营养组,180 d>120 d>90 d),差异有统计学意义(P<0.05或0.01);与对照组比较,各染毒组ABP和Tf表达均减少,且随染毒剂量的增加和时间的延长,表达愈加减少(P<0.05或P<0.01);在180 d时,同阶段同剂量染毒大鼠,加营养组ABP和Tf表达水平较高(P<0.05)。结论:在燃煤型氟中毒大鼠睾丸中,随染毒剂量增加和染毒时间延长,ABP和Tf表达均减少;降低摄氟量及改善营养状况,可改善燃煤型氟中毒对大鼠睾丸ABP和Tf表达的影响。Objective: To study effects of coal-burning fluorosis on expression of androgen binding protein( ABP) and transferrin( Tf) in rat testis. Methods: The male SD rats were randomly divided into 7 groups,including control group,low-dose fluoride group,middle-dose fluoride group,high-dose fluoride group,low-dose fluoride supplemented with nutrition group,middle-dose fluoride supplemented with nutrition group and high-dose fluoride supplemented with nutrition group. The experimental groups were fed with the fodder with different proportions of corn dried by burning coal coming from endemic fluorosis areas to establish the animal model of coal-burning fluorosis( soybean flour was added in nutrition-involved groups). All rats were killed by femoral arterial exsanguination at three times( 90 d,120 d,180 d),followed by examinations of dental fluorosis,fluorine in urine. Immunohistochemical method was adopted to observe and analyze the changes of expression of ABP and Tf in rat testis. Results: The severity of dental fluorosis and the fluorine content in urine increased gradually along with the increase of exposure dose and time,and the differences between each group were statistically significant( P〈0. 05 or 0. 01). Compared with the control groups,ABP and Tf expression in other experimental groups decreased significantly and with the increase of exposure dose and time,ABP and Tf expression decreased further( P〈0. 05 or 0. 01). At day 180,compared with other groups of the same exposure dose and time,ABP and Tf expression level was relatively higher. Conclusions: With the exposure dose and exposure time extended,ABP and Tf expressed decreasingly in coal-burning fluorosis rat testis. Reducing fluorine intake and improving nutritional status could improve the expression of ABP and Tf in coal-burning fluorosis rat testis.
