大鼠心脏移植急性排斥反应与CD8^+CD28^+T淋巴细胞钙离子的相关性研究

摘　　要：目的探讨CD8^+CD28^+T淋巴细胞胞外^(45)Ca^(2^+)摄取率、胞内Ca^(2^+)浓度与大鼠心脏移植急性排斥反应(AR)的相关性。方法用流式细胞仪分选大鼠对照组(T0)、假手术组(T_1)、实验组(T_2)术后第1、3、5、7、9、11天外周血CD8^+CD28^+T淋巴细胞,测量胞外^(45)Ca^(2^+)摄取率、胞内Ca^(2^+)浓度,进而分析CD8^+CD28^+T淋巴细胞胞外^(45)Ca^(2^+)摄取率、胞内Ca^(2^+)浓度与大鼠心脏移植后AR的相关性。结果T2组CD8^+CD28^+T淋巴细胞胞外^(45)Ca^(2^+)摄取率、胞内Ca^(2^+)浓度较T_1、T_0组明显升高,差异有统计学意义(P<0.05);与心脏移植后AR程度的Spearman相关系数分别为0.494(P<0.01)、0.688(P<0.01)。结论外周血CD8^+CD28^+T淋巴细胞胞外^(45)Ca^(2^+)摄取率、胞内Ca^(2^+)浓度与大鼠心脏移植AR密切相关,有助于早期监测心脏移植术后AR。Objective To explore the correlation between the acute rejection（AR）in rat heart transplant（HT）with extracellular 45Ca2＋uptake rate and intracellular Ca2＋concentration of CD8〈sup〉＋CD28〈sup〉＋T lymphocytes.Methods The flow cytometry was used to sort the peripheral blood CD8＋CD28＋T lymphocytes on postoperative 1,3,5,7,9,11 din the rat control group（T0）,sham operation group（T1）and experiment group（T2）,the extracellular45Ca2＋uptake rate and intracellular Ca2＋concentration were measured,and then the correlation between extracellular 45Ca2＋ uptake rate and intracellular Ca2＋concentration of CD8＋CD28＋T lymphocytes was analyzed.Results The extracellular 45Ca2＋uptake rate and intracellular Ca2＋concentration of CD8＋CD28＋T lymphocytes in the T2 group were significantly increased compared with the T1 group and T0 group,the difference was statistically significant（P〈0.05）;the 45Ca2＋uptake rate and intracellular Ca2＋concentration of CD8＋CD28＋T lymphocytes in the T2 group were positively correlated with the degree of AR,the Spearman correlation coefficients were 0.494 and 0.688respectively（P〈0.01）.Conclusion Peripheral blood extracellular 45Ca2＋uptake rate and intracellular Ca2＋concentration of CD8＋CD28＋T lymphocytes are closely correlated with the AR in rat HT,which is conducive to early monitor AR after HT.
