In order to identify the effects of biochar on carbon and nitrogen metabolism of flue-cured tobacco, taking the tobacco variety( Nicotianatabacum L,K326) as material, conducted the systematic research from the cell tissue structure, gene expression and metabolism in three levels. Results showed that the starch were more at the earlier stages, and degraded more completely in the treatment of no applying biochar with leaf maturity. The analysis of gene expression displayed that the expression of the genes related to carbon metabolism increased gradually after topping, while the expression of the genes related to nitrogen metabolism reduced. The expression of the genes were higher by applying more biochar at the later stages. The analysis of substance contents indicated that the contents of total sugar and reducing sugar reduced after applying biochar, the contents were higher after cured. The starch contents were higher after applying 600 kg / hm2. The contents of total nitrogen and nicotine increased after applying biochar, the difference was diminished after cured, these results showed that the metabolism of tobacco carbon and nitrogen were affected by applying biochar, thus affected the quality of flue-cured tobacco.
