摘 要:为了探明有机肥用量对烤烟碳氮代谢途径关键基因表达的影响,设置3种有机肥施用量(0、200、600 kg/667m2)来分析烤烟成熟期叶片碳氮代谢相关关键基因的表达水平。结果表明:施用有机肥显著降低了碳代谢途径中蔗糖合成酶基因(SUS)、蔗糖磷酸合成酶基因(SPS)、颗粒结合型淀粉合成酶基因(GBSSI)和淀粉分支酶基因(SBE)的表达水平,3个处理表现为:CK>C200>C600。通过对氮代谢途径关键基因硝酸还原酶(NITR)、亚硝酸还原酶(NIR)、谷氨酸脱氢酶(GDH)和谷氨酰胺合成酶(GS)中一个重要家族成员(GLN1-3)的表达分析表明:施用适量(200 kg/667 m2)有机肥对氮代谢途径关键基因的表达量影响不显著,仅在高用量(600 kg/667 m2)有机肥条件下表现出抑制。因此,通过添加不同有机肥用量对烤烟碳氮代谢关键基因的表达分析可以看出,有机肥处理下的碳氮代谢基因表达量明显受到抑制,且与有机肥的施用量呈正相关性,有机肥对碳代谢途径的影响大于氮代谢。In order to determine the effects of organic fertilizer application on carbon and nitrogen metabolism key genes expressions of flue-cured tobacco,different organic fertilizer applications( 0,200,600 kg / 667 m2) were set in this study,which analyzed carbon and nitrogen metabolism related genes expressions of flue-cured tobacco in maturity. The results showed that sucrose synthase( SUS),sucrose phosphate synthase( SPS),granule- bound starch synthase( GBSSI) and starch branching enzyme( SBE) were decreased conspicuously by applying organic fertilizer and three treatments were as follows: CKC200 C600.The expressions of nitrogen metabolism related genes nitrate reductase( NITR),nitrite reductase( NIR),glutamate dehydrogenase( GDH) and an important family members( GLN1-3) of glutamine synthesis enzymes( GS) were elucidated that nitrogen metabolism related genes expressions were not significant in applying appropriate organic fertilizer( 200 kg / 667 m2) and inhibited in higher organic fertilizer( 600 kg / 667 m2). Therefore,which could be found that expressions of carbon and nitrogen related gene of flue-cured tobacco were inhibited significantly and positive correlation with organic fertilizer application by applying different organic fertilizer,the effect of organic fertilizer on carbon metabolism was more than that of nitrogen metabolism.
