仙灵骨葆胶囊含药血清对兔软骨细胞增殖及分泌功能的影响

摘　　要：目的:通过体外软骨细胞培养的方法,观察仙灵骨葆胶囊含药血清对新西兰兔膝软骨细胞增殖凋亡及糖胺聚糖(GAG)分泌的影响。方法:将新西兰大白兔后肢膝关节软骨细胞分离培养,化学染色鉴定,用4组不同浓度仙灵骨葆胶囊含药血清干预分离软骨细胞,观察其形态学,观察软骨细胞及MTT检测细胞增殖率,邻联甲苯胺(DMB)染色法检测细胞内GAG含量。结果:空白血清组及不同剂量含药血清组均使兔软骨细胞大量增殖(P<0.01),且强弱依次为低浓度组>中浓度组>空白组>高浓度组>对照组;空白血清组及不同浓度的含药血清组均使兔软骨细胞GAG的合成增多(P<0.01),且强弱依次为高浓度组>低浓度组>中浓度组>空白组>对照组。结论:仙灵骨葆胶囊各浓度含药血清可不同程度地刺激兔软骨细胞增殖,分泌GAG,低浓度综合作用最强。Objective： By culturing the chondrocytes in vitro, to observe the effects of serum containing Xianling Gubao Capsule on proliferation and apoptosis of rabbit knee cartilage cell and the secretion of glycosaminoglycan（GAG）. Methods： The chondrocytes from the hind knee joint of New Zealand rabbit were incubated, and the content was identified by toluidine blue stains and immunohistochemical ABC methods. Then the chondrocytes were cultured with 4 different concentrations of serum containing Xianling Gubao Capsule groups. The morphological of chondrocytes and the MTT rate were observed, and the content of intracellular GAG was detected by o-tolidine（DMB） staining. Results： The proliferation of rabbit chondrocytes in blank serum group and different concentrations of drug serum groups were increased（P〈0.01）, and the order of proliferation from high to low was low concentration group, middle concentration group, blank group, high concentration group and control group; the GAG of rabbit articular chondrocytes in the blank serum group and different concentrations of drug serum groups increased（P〈0.01）, and the order from high to low was high concentration group, low concentration group, middle concentration group, blank group, control group. Conclusion： The different concentrations serum containing Xianling Gubao Capsule could stimulate the proliferation and secretion of rabbit chondrocytes, but the low concentration has the strongest effect.
