从L型钙通道研究益气养阴复方治疗早搏的作用机制

摘　　要：目的:通过观察益气养阴复方治疗早搏大鼠,研究作用机制,为临床治疗提供依据。方法:选取120只大鼠,20只作为正常组,其余100只大鼠采用舌下静脉注射乌头碱配合噪音建立慢性早搏大鼠模型,将造模成功大鼠随机分成5组,即模型组、中药大剂量组、中药中剂量组、中药小剂量组及西药组。4周后,观察各组大鼠精神状态、毛色、口唇、舌质、呼吸和心率等一般情况变化及心肌组织L-型钙通道基因和Ca^(2+)-ATP酶基因的表达情况。结果:较正常组相比,模型组、中药高、中、低剂量组和西药组大鼠LCCs和ca^(2+)-ATP酶水平下降(P<0.05);与模型组比较,中药高、中、低剂量组和西药组大鼠LCCs和ca^(2+)-ATP酶水平升高(P<0.05),且中药中、高剂量组优于中药低剂量组(P<0.05),中药中、高剂量组提高ca^(2+)-ATP酶基因表达优于西药组(P<0.05);中药高、中、低剂量组和西药组均能够明显改善房颤大鼠的精神状态、毛色、口唇、舌质、呼吸和心率等一般状态。结论:益气养阴复方能够提高早搏大鼠LCCs和ca^(2+)-ATP酶的表达,作用机制可能与L型钙离子通道的信号通路有关,对心律失常的预防与治疗有着广阔前景。Objective： Through the observation of Yiqi Yangyin recipe in treating premature rats, mechanism of tra- ditional Chinese medicine treatment of premature beat, for clinical treatment provides the basis. Methods： 120 rats and 20 as the normal group, the remaining 100 rats by sublingual venous injection of aconitine to establish a rat model of chronic beats with the noise, the successful model rats were randomly divided into 5 groups： model group, high dose of Chinese medicine group, Chinese medicine dose group, traditional Chinese medicine small dose group and Western medicine group. After 4 weeks, and the mental state of each group rats was observed, the coat color of the lips, tongue, breathing and heart rate in general and the changes of myocardium L-type calcium channel gene and Ca2＋-ATPase gene expression. Results： Compared with normal group, model group, traditional Chinese medicine, high and low dose group and Western medicine group rats of LCCs and Ca2＋ - ATP enzyme levels decreased （P〈0.05）; and the model group, herbal therapy high, middle, low dose group and Western medicine group rats of LCCs and Ca2 ＋ - ATP enzyme levels are elevated （P〈0.05）, and the traditional Chinese medicine, high dose group was better than that of traditional Chinese medicine （TCM） low dose group （P〈0.05） in traditional Chinese medicine, high dose group increased Ca2＋-ATP enzyme gene expression is superior to western medicine group （P〈0.05）; traditional Chinese medicine high, medium and low dose group and Western medicine group were able to significantly improve the atrial fibrillation mental state,hair color, lip, tongue, breathing and heart rate in general. Conclusion： Yiqi Yangyin recipe can increase the expression of premature rats of LCCs and Ca2 ＋-ATPase, related signal pathway in the mechanism of action may and L type calcium ion channel and the prevention and treatment of arrhythmia with broad prospects.
