摘 要:目的探讨丹参川芎嗪注射液对缺氧条件下脑基底平滑肌细胞的保护作用,并比较丹参素、川芎嗪、丹参川芎嗪注射液三者的药效差异。方法用Na_2S_2O_4与细胞共孵育,建立大鼠脑基底平滑肌细胞缺氧/复氧损伤模型。对照组换上新鲜培养基;模型组给予缺氧液;丹参川芎嗪低、中、高浓度组分别给予0.01,0.1,1μmol·L^(-1)丹参川芎嗪注射液孵育6 h;丹参素组给予0.1μmol·L^(-1)丹参素孵育6 h;川芎嗪组给予0.1μmol·L^(-1)川芎嗪孵育6 h;丹参素+川芎嗪组给予0.1μmol·L^(-1)丹参素+0.1μmol·L^(-1)川芎嗪孵育6 h。观察脑基底平滑肌细胞一般形态学及凋亡相关形态学的改变、检测脑基底平滑肌细胞存活率、脑基底平滑肌细胞外液乳酸脱氢酶活性、以及Bcl-2/Bax的表达。结果与模型组相比较,给药组脑基底平滑肌细胞的存活率上升(P<0.05或0.01),Bcl-2/Bax的比值升高(P<0.05),降低了低氧损伤引起的细胞外液乳酸脱氢酶的升高(P<0.05)。结论丹参川芎嗪注射液对缺氧造成的脑基底平滑肌细胞损伤具有保护作用,其保护作用优于丹参素、川芎嗪。Objective To explore salvia miltiorrhizaligustrazine injection on cerebral basal smooth muscle cells under hypoxia ischemia and compare the efficacy of salvia miltiorrhiza,ligustrazine and salvia miltiorrhizaligustrazine injection. Methods Select cerebral basal smooth muscle cells to study. Use of sodium dithionite( Na2S2O4) were incubated with the cells caused by oxygen and glucose deprivation injury. The experiment will be divided into control group( fresh medium),model group( a lack of oxygen liquid),salvia miltiorrhizaligustrazine low,middle,high-dose groups( 0. 01,0. 1,1 μmol·L^-1,6 h),salvia miltiorrhiza group( 0. 1 μmol·L^-1,6 h),ligustrazine group( 0. 1 μmol·L^-1,6 h),salvia miltiorrhiza plus ligustrazine group( 0. 1 μmol · L^-1,6 h).Observe the general morphology of cerebral basal smooth muscle cells and morphological changes of apoptosis related,detection of cerebral basal smooth muscle cells survival,cerebral basal smooth muscle cells activity of extracellular lactate dehydrogenase( LDH) and the activity of Bcl- 2 / Bax expression. Results Salvia miltiorrhizaligustrazine injection reduce hypoxia- reperfusion injury of cerebral basal smooth muscle cells in the general form. Compared with model group,it improve the survival rate ofcerebral basal smooth muscle cells( P〈0. 05 or 0. 01),increase the ratio of Bcl- 2 / Bax( P〈0. 05),and decrease the oxygen damage caused by extracellular LDH increased( P〈0. 05). Conclusion Salvia miltiorrhizaligustrazine injection has protective effect on the injury of cerebral basal smooth muscle cells induced by ischemia and hypoxia. Its protective effect superior to that with salvia miltiorrhiza andligustrazine.
