不同温补肾阳中药主要组分对小鼠肾上腺皮质瘤细胞增殖及皮质酮分泌的影响

摘　　要：目的：研究温补肾阳中药主要组分对小鼠肾上腺皮质瘤细胞增殖及皮质酮分泌的影响。方法：以Y1小鼠肾上腺皮质瘤细胞为实验对象,分别采用0.2~640μmol/L淫羊藿苷、仙茅苷、金丝桃苷、松果菊苷、松脂醇二葡萄糖苷、补骨脂素和耐斯糖干预Y1细胞24~72 h;并以0.1%DMSO为对照组,佛司可林为诱导剂组;通过MTT法检测细胞增殖、q PCR检测细胞皮质酮合成酶基因表达、ELISA检测细胞上清液皮质酮水平。结果：Y1细胞铺板0~72 h内,对照组细胞增殖明显,80~640μmol/L淫羊藿苷治疗72 h显著抑制Y1细胞增殖,且呈现量效关系（P〈0.05）;640μmol/L松果菊苷治疗24 h、48 h和72 h均显著抑制Y1细胞增殖（P〈0.05）;10~80μmol/L补骨脂素治疗48 h显著促进Y1细胞增殖（P〈0.05）;大于320μmol/L补骨脂素治疗72 h显著抑制Y1细胞增殖（P〈0.05）;然而,仙茅苷、金丝桃苷、松脂醇二葡萄糖苷、耐斯糖对Y1细胞增殖无明显影响。基础状态下,与对照组比较,仅10μmol/L补骨脂素显著增强Cyp11b1基因表达（P〈0.05）;佛司可林显著促进各组Star、Cyp11a1和Cyp11b1基因表达及皮质酮分泌（P〈0.05）。结论：淫羊藿苷、松果菊苷和补骨脂素对Y1细胞增殖抑制作用与其浓度和作用时间有关,本研究中采用的温补肾阳中药主要组分对Y1细胞皮质酮合成与分泌作用弱。Objective： To investigate the effects of different warming the kidney-yang Chinese medicine on Y1 adrenocortical cell proliferation and secretion of corticosterone. Methods： Y1 adrenocortical cell were choosed and 0. 2 ~ 640μmol /L Icariin,Curculigoside,Hyperoside,Echinacoside,Pinoresinol diglucoside,Psoralen,Nystose were used respectively to intervened Y1 adrenocortical cell 24 ~ 72 h; 0. 1% DMSO was treated as control group and Forskolin as Revulsant group;the cell proliferation was detected by MTT,gene expression of cells corticosterone synthetase was detected by q PCR,level of corticosterone in the supernatant fluid was detected by ELISA. Results： Y1 adrenocortical cells were planked 0 ~ 72 h,there was obvious proliferation in control group,Y1 adrenocortical cells were obviously inhibited with treatment of 80 ~ 640 μmol / L Icariin for 72 h in a dose-dependent manner（ P 0. 05）; Y1 adrenocortical cells all were obviously inhibited with treatment of640 μmol / L Echinacoside for 24 h,48 h and 72 h（ P 0. 05）; Y1 adrenocortical cells were obviously promoted with treatment of 10 ~ 80 μmol / L Psoralen for 48h（ P 0. 05）; Y1 adrenocortical cells were obviously inhibited with treatment of over 320μmol /L Psoralen for 72h（ P 0. 05）; while there was no obvious effect with treatment of Curculigoside,Hyperoside,Pinoresinol diglucoside and Nystose. Compared with control group,only 10 μmol / L Psoralen could promote the expression of Cyp11b1 on the elemental status（ P 0. 05）; Forskolin could promote the expression of Star,Cyp11a1 and Cyp11b1 and secretion of corticosterone（ P 0. 05）. Conclusion： The inhibitory effect of Icariin,Echinacoside and Psoralen on Y1 adrenocortical cells had the relationship with concentration and action time,and there was little effect of warming the kidneyyang Chinese medicine on Y1 adrenocortical cell proliferation and secretion of corticosterone in this study.
