摘 要:目的探讨N-乙酰半胱氨酸(NAC)对常见呼吸道感染病原菌的抑菌作用及其影响抑菌作用的因素。方法以常见呼吸道感染病原菌为供试菌,通过有限稀释法和悬液定量杀菌法进行抑菌试验。观察不同影响因素(不同浓度、不同时间、p H、有机物质和存放时间)对NAC抑菌作用的影响。结果 NAC对金黄色葡萄球菌最小抑菌浓度(MIC)为2.5mg/ml,最小杀菌浓度(MBC)为5mg/ml。对大肠埃希菌和肺炎克雷伯菌的MIC为5mg/ml,MBC为10mg/ml。悬液定量杀菌结果表明,5mg/ml NAC,在菌终浓度约为104cfu/ml,作用40min,对金黄色葡萄球菌、大肠埃希菌、铜绿假单胞菌、肺炎克雷伯菌、嗜食麦窄食单胞菌、肺炎链球菌和乙型溶血型链球菌,杀菌率几乎达到100%;p H=5.4~7.5之间,在作用40min,20mg/ml的NAC对金黄色葡萄球菌、大肠埃希菌的杀菌作用与相应p H浓度的PBS之间无显著差异;p H=4.0时,NAC具有明显的杀菌作用,而相同的PBS无明显杀菌作用;有机物质(小牛血清)8%以上时,NAC对金黄色葡萄球菌、大肠埃希菌杀菌作用明显降低;在密封条件下,冷藏3d,NAC仍能保持较好的杀菌活性,但室温条件存放1d以上,杀菌效果就会有所降低。结论 NAC对常见呼吸道感染菌具有较好的抑菌效果;环境中的有机物质、酸碱度是影响NAC抑菌效果的主要因素。Objective To explore the bacteriostasis effect of N- acetyl cysteine( NAC) on common respiratory pathogens and its influencing factors. Methods Limited dilution method and suspension quantitative germicidal test were tested,and common respiratory tract infection pathogens were used on the bacteriostatic experiments. It was observed by different influence factors( different concentration,different time,p H,organic matter,storage time) on bacteriostasis effect of the NAC. The plate pouring method was used the colony forming unit( CFU). Results The minimal inhibitory concentration( MIC) of NAC was 2. 5 mg / ml for Staphylococcus aureus,while the minimum bactericidal concentration( MBC) was 5mg / ml. The MIC was 5 mg / ml for Escherichia coli and Klebsiella pneumonia,while the MBC was 10 mg / ml. By suspension quantitative germicidal method,the sterilization rate was up to 100%,when the bacteria were exposed to 5mg / ml NAC for 40 min at the concentration of 104 CFU /ml for S. aureus,E. coli,K. pneumoniae,S. maltophilia,S. pneumoniae,and S. hemolytic- β. When it was used by the Suspension quantitative germicidal test,the bactericidal action was no significant difference between 20 mg / ml of NAC and PBS at the condition of p H = 5. 4- 7. 5,for 40 min on S. aureus and E. coli. The bacteriostasis was an obvious difference under p H = 4. 0,20 mg / ml of NAC,but it was no significant differences in the same PBS. The bacteriostasis of NAC was decreased obviously under the organic matter( calf serum) more than 8% for 40 min on S. aureus and E. coli. In an airtight condition,the refrigeration3 d,NAC can still maintain good bactericidal activity,but at room temperature for more than 1d,the bactericidal effect could be greatly reduced. Conclusion The bacteriostatic effect of NAC on respiratory infection pathogens is good. The organic matter and p H in the environment are the main factors affecting the bacteriostatic effect of NAC.
