摘 要:目的建立测定胡颓子叶中总黄酮及槲皮素、山柰酚和异鼠李素含量的方法。方法采用紫外分光光度法,以芦丁为对照品,以三氯化铝(Al Cl3)显色,在411 nm处测定胡颓子叶中总黄酮的含量。采用水解-HPLC法同时测定胡颓子叶中槲皮素、山柰酚和异鼠李素的含量。色谱条件采用Ultimate KAQ-C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸梯度洗脱,流速为1.0 ml/min,检测波长为360 nm,柱温为30℃。结果芦丁的质量浓度在0.0056~0.028mg/ml与吸光度呈良好线性关系(r=0.9997),平均加样回收率为100.99%,RSD=1.89%(n=9)。槲皮素、山柰酚、异鼠李素3种黄酮苷元质量浓度分别在9.91~59.46μg/ml(r=0.9999)、46.6~372.8μg/ml(r=0.9998)、0.99~19.8μg/ml(r=0.9998)与峰面积积分值呈良好的线性关系,平均加样回收率分别为101.87%,101.21%,97.62%(RSD均小于2%,n=6)。胡颓子叶样品中总黄酮、槲皮素、山柰酚和异鼠李素的含量分别为11.94,0.34,1.89,0.03 mg/g。结论该方法简便可靠,重复性好,可作为胡颓子叶中黄酮类物质的含量测定方法。Objective To establish the method for the content determination of total flavonoids and quercetin,kanpferol and isorhamnetin in Elaeagnus pungens. Methods Using rutin as control,the content of total flavonoids in Elaeagnus pungens was determined by UV spectrometry. The contents of quercetin,kaempferol and isorhamnetin were determined by HPLC after hydrolysis.The analysis was carried out on an UltimateAQ-C18column( 250 mm × 4. 6 mm,5 μm) and detected with an UV detector at the wavelength of 360 nm. Gradient elution was carried out with methanol-0. 1% phosphoric acid at the flow rate of 1. 0 m L /min. The column temperature was set at 30 ℃. Results The linear range was 0. 0056 ~ 0. 028 mg / m L for rutin( r = 0. 9997)with an average recovery of 100. 99%( RSD = 1. 89%,n = 9). The linear range of quercetin,kaempferol and isorhamnetin was9. 91 ~ 59. 46 μg / ml( r = 0. 9999),46. 6 ~ 372. 80 μg / ml( r = 0. 9998) and 0. 99 ~ 19. 8 μg / m L( r = 0. 9998),respectively.The average recoveries of the three components were 101. 87 %,101. 21 % and 97. 62 %,respectively. The total flavonoids and3 maker compounds in Elaeagnus pungens were 11. 94,0. 34,1. 89,0. 03 mg / g,respectively. Conclusion The method is simple,reliable and repeatable,and can be used for the simultaneous determination of flavonoids in Elaeagnus pungens.
