摘 要:目的探讨左归降糖解郁方对糖尿病并发抑郁症模型大鼠海马NR不同亚型NR2A、NR2B表达的影响。方法建立糖尿病并发抑郁症大鼠模型,并随机分为5组:模型组,阳性药(二甲双胍0.18g/kg+盐酸氟西汀1.8mg/kg)组,左归降糖解郁方高、中、低剂量(32.82,16.41,8.20g/kg)组,以正常大鼠为空白对照组,每组10只,灌胃给药28天。给药后,采用Morris水迷宫实验评价大鼠行为学变化,Western-blotting和RT-PCR法定量检测各组大鼠海马NR2A、NR2B相关基因及蛋白的表达情况。结果与空白对照组比较,模型组大鼠(P<0.01)逃避潜伏期延长,空间探索时间减少,海马NR2A、NR2B表达明显升高(P<0.01),差异有显著统计学意义;与模型组比较,阳性药组和左归降糖解郁方高剂量组大鼠逃避潜伏期明显缩短,空间探索时间增加(P<0.01),海马NR2A、NR2B的表达减少(P<0.05),差异有统计学意义。结论左归降糖解郁方可通过调控DD大鼠海马NR2A、NR2B的表达,明显改善DD大鼠的抑郁样行为。Objective To investigate the effect of Zuo-gui Jiang-tang Jie-yu Fang( ZJJF) on NR2 A and NR2 B expression in hippocampus in diabetes mellitus rats with depression( DD). Methods DD rat model was established,the model rats were randomly divided into five groups,the DD group,metformin( 0. 18 g / kg) combined fluoxetine( 1. 8 mg / kg) group,high-,mid-,and low-dose( 32. 82,16. 41,and8. 20 g / kg) ZJJF groups. Normal rats act as control group,10 rats per group. After 28 days of administration,Morris test were used to detect the behavior of the rats,the expression of NR2 A,NR2B in rat hippocampus was detected by Western-blotting and RT-PCR. Results Compared with control group,the escape latencies were significantly longer in the Morris water maze test( P〈0. 01),as well as the space exploration times were significantly shortened( P〈0. 01).and NR2 A,NR2B expression( P〈0. 01) were increased. Compared with the model group,the escape latencies were significantly longer in the Morris water maze test( P〈0. 01),as well as the space exploration times were significantly shortened( P〈0. 05),NR2 A,NR2B expression was decreased( P〈0. 05). Conclusion ZJJF improved depressive behavior of diabetic rats with depression may be related to the regulation of NR2 A and NR2 B expression in hippocampus.
