摘  要:目的分析骨髓机制干细胞在兔DBM上增殖作用。方法 2014年3月—2015年7月,以3只大白兔为研究对象,根据Urist方法制备出DBM(同种脱钙骨基质),测定光密度(OD)值,绘制增殖曲线,分析复合培养后最佳修复软骨移植时机。结果 SEM观察呈现高密度空隙网架结构,平均孔隙率为(60.19±6.1)%;平均孔径大小为(119.42±6.46)μm;兔DBM材料BMSCs体外复合培养后可见大量骨髓基质干细胞粘附,MTT法检测复合培养6d骨髓基质干细胞增殖达到稳定状态。结论兔脱钙骨基质具有三维网状结构和良好的细胞相容性是一种较好的软骨组织工程支架材料,最适合移植。Objective To analyze the value-added function of bone marrow stromal stem cells in rabbit DBM. Methods From March 2014 to July 2015, 3 big white rabbits were selected as the research object, DBM(demineralized allogeneic bone matrix) was prepared according to the Urist method, the optical density(OD) value was detected, the value-added curve was drawn and the best repair cartilage transplantation after compound culture was analyzed. Results High density space truss structure was presented by SEM observation, the average porosity was(60.19±6.1)% and the average pore diameter size was(119.42±6.46) μm, a large number of bone marrow stromal stem cells adhesion was seen on the rabbit DBM materials after BMSCs compound culture in vitro, the value – added of bone marrow stromal stem cells ?reached a steady state after 6d of compound culture by the detection of MTT method. Conclusion Rabbit DBM has a three-dimensional network structure and good cell compatibility, which is a good cartilage tissue engineering scaffold material and most suitable for transplantation.
