摘  要:目的建立HPLC法,用于测定患者体内血浆中胺碘酮及其代谢产物去乙基胺碘酮血药浓度。方法采用色谱柱Ultimate C18(250 mm×4.6 mm,5μm)柱;流动相:醋酸-三乙胺缓冲溶液∶甲醇=19∶81;检测波长242 nm;流速1.0 m L·min-1,以黄体酮为内标。结果胺碘酮线性关系为y=0.227 7 x-0.000 1,r=0.999 5;去乙基胺碘酮线性关系y=0.220 6x-0.015 6,r=0.999 8,回收率>90%、RSD<10%。结论此方法简便、准确,可操作性强,可快速准确地检测出患者的胺碘酮及去乙基胺碘酮血药浓度,值得推广使用。AIM To build a method with HPLC to determine the blood concentration of amiodarone and its metabolite-desethylamidarone from patients. METHODS The column was Ultimate C18 (250 mm × 4.6 mm, 5 μm)column. Mobile phase was glacial acetic acid-triethylamine buffer solution:methanol = 19:81; The detection wavelength was 242 nm; Progestrone was used as the internal standard. RESULTS The regression equation of amiodarone was y = 0.227 7 x -0.000 1, r = 0.999 5; The regression equation of desethylamiodarone was y = 0.220 6 x -0.015 6, r = 0.999 8 .The recovery rate was 〉 90%, and the RSD was 〈 10%. CONCLUSION This method is simple and accu- rate, it can quickly determine the blood concentration of amiodarone and desethylamiddarone in patients' plasma.
