摘 要:目的:观察黄连素对实验性糖尿病大鼠肝糖原的影响,探讨黄连素的降血糖作用机制。方法:采用链脲佐菌素尾静脉注射复制实验性糖尿病模型,实验分为正常组、模型组、黄连素低剂量组、黄连素高剂量组,每组10只。分组给药4周后,测定大鼠的空腹血糖、肝糖原含量、糖异生转化率、肝脏葡萄糖激酶(GK),观察肝糖原PAS染色结果。结果:黄连素明显降低实验性糖尿病大鼠空腹血糖,抑制糖异生,降低糖异生转化率,增强GK活性,增加肝糖原含量,增强肝糖原PAS染色。结论:黄连素对实验性糖尿病大鼠肝糖原的影响可能通过抑制糖异生,增强GK活性来实现的。Objective: To investigate the mechanism of Berberine on lowering blood sugar by observing the effect of Berberine on hepatic glycogen of diabetes rats. Methods: Diabetic models were made by injecting STZ, experiment was divided into 4 groups ,normal ,model, berberine high ,low dose groups, 10 rats in each group,they were given medicine or NS for 4 weeks. The fasting plasma glucose, glycogen, gluconeogenesis conversion rate, liver glucokinase (GK)activity, PAS staining of glycogen were datermined. Results: Berberine could signifi- cantly reduce fasting glucose, inhibite gluconeogenesis, reduce gluconeogenesis conversion rate, and enhance GK activity, increase glycogen content and enhance glycogen PAS stain in diabetic rats. Conclusion:Effects of Barberine on glycogen of experimental diabetic rats may be related to inhibiting gluconeogenesis and enhance GK activity.
