摘  要:目的:观察护卵颗粒对重复制动应激致大鼠卵巢功能减退的治疗作用。方法:模型组采用性周期规律的雌性SD大鼠置于特制的束缚制动筒内,固定身体使之不能随意活动,制动应激1次/d,连续15 d建立卵巢早衰大鼠模型,每天阴道图片观察大鼠动情周期变化,造模成功后分为6组:模型对照组、补佳乐组、调经促孕丸组及护卵颗粒高、中、低剂量组,另取10只作为空白对照组,连续给药4周。给药结束后经开野行为测定后腹主动脉采血,冰台上取大鼠下丘脑,留取卵巢组织标本。测定血清雌二醇(Estradiol in serum,E2)、促卵泡激素(Follicle-stimulating Hormone,FSH)、黄体生成素(Luteinizing Hormone LH)、孕酮(Progesterone,PROG)水平及检测下丘脑多巴胺(Dopamine,DA)、去甲肾上腺素(Noradrenaline,NA)、五羟色胺(Hydroxytryptamine,5-HT)含量,观察卵巢湿重变化及组织学改变。结果:护卵颗粒各剂量组能明显提高重复制动应激致卵巢早衰大鼠的水平和垂直活动得分,能提高卵巢早衰大鼠血清E2、PROG水平和降低FSH、LH水平,降低卵巢早衰大鼠下丘脑DA、NA及5-HT含量,与模型组比较差异均有统计学意义(P<0.05或P<0.01)。与模型组比较,护卵颗粒高、中剂量组大鼠卵巢内各级卵泡数增多,卵巢指数增加。结论:护卵颗粒可以提高卵巢功能减退模型大鼠水平活动和垂直活动,增加E2、PROG及NA的分泌,增加卵巢指数,促进卵巢功能减退大鼠的卵泡生长,恢复受损卵巢的内分泌功能。Objective: To observe the effects of Hu Luan Ke Li on diminished ovarian reserve in rats of repeated immobilization. Methods: The adult female rats with regular cycle in model group were exposed to the scheduled daily repeated immobilization for 15 days,one time per day. The duration of immobilization was progressively increased with the subsequent episode. Daily vaginal smears were permfored to follow the estrous cycle in rats. After molding,60 rats were subdivided into 6 groups( n= 10) : Model group,Bu Jia Le group,Tiao Jing Cu Yun Wan group,high-dose、middle-dose and low-dose group,treated for 4 weeks respectively. After Openfield testing,blood samples were obtained from abdominalaorta,and samples of ovarian tissues were harvested. Serum levels of Estradiol( E2),Follicle-stmiulating hormone( FSH),Luteinizing hormone( LH),Progesterone( PROG) and hypothalamus content of Dopamine( DA),Norepinephrine( NA),Serotonin( 5-HT) were measured by ELISA,and the changes of wet weight and histology of ovary were observed. Results: Compared with the model group,Hu Luan Ke Li increased serum level of E2,PROG and decreased FSH,LH; and decreased hypothalamus content of DA,NA,5-HT( P〈0. 05 or P〈0. 01); and the wet weight of ovary increased in each dose of Hu Luan Ke Li groups. Conclusion: Hu Luan Ke Li can promote follicle growth in rats with premature ovarian failure,increase secretion of ovarian E2,PROG,and improve endocrine function of damaged ovaries.
