水曲柳复幼——微繁技术研究

摘  要:采用截干、喷施激素、掩埋锯末三种处理方式对水曲柳2年生幼苗进行复幼处理,并以萌生枝条的顶芽及腋芽作为外植体,探讨最佳复幼——微繁体系,结果表明:(1)截干+喷施激素处理的幼苗产生的穗条数量、长度及直径均优于其它处理,每株幼苗当年可产出6~7条可供嫁接的穗条和20个左右茎节外植体(水曲柳对照平均为4~5个)。(2)复幼处理获得的萌条茎节进行组织培养时,新芽启动率和增殖系数均超出对照,其中截干+喷施BA组合最为突出,启动率超出对照的132%,而截干+喷施激素+掩埋锯末初代增殖系数最高,超出对照65.2%。(3)经隶属函数评分排序,选出附加ZT 1.0 mg/L和2%蔗糖的WPM培养基最有利于新芽分化,增殖系数达到9.61,与其它处理差异显著。2 years old seedlings of Fraxinus mandschurica were treated by cutting, spraying hormones and covering with sawdust. In order to explore the influence of different treatments on the seedlings, the apical and lateral buds that germinated from the treated shoots were cultured in the same medium. And then the culture condition of tissue culture seedlings was optimized by using the same medium with different hormones combinations.The results showed that:(1) the number, length and diameter of coppice shoot that came from young seedlings and treated by cutting and hormones, were better than other treatments. Under this treatment, one seedling could produce six to seven coppice shoots for grafting or about twenty stems for tissue culturing every year.(2) The average germinate rate of explants and the propagation coefficient of the tissue culture seedlings were higher than the untreated seedlings. The germinate rate of the explants which came from cutted and sprayed seedlings was 232% of CK. But the propagation coefficient of the tissue culture seedlings which came from seedlings treated with three treatments was 165% of CK.(3)As to the attaching functional value,the tissue culture seedlings from four different medium were evaluated, the WPM medium with 2% sugar and 1.0mg/L ZT were best for seedlings propagation. The propagation coefficient was as high as 9.61,which reached significant level comparing with other treatments.
