摘 要:卵泡颗粒细胞对卵母细胞的营养支持、保护以及卵泡生长发育、成熟、闭锁、维持黄体功能等起着至关重要的作用。为研究湖羊黄体期卵泡颗粒细胞的基因表达及其信号网络,提取湖羊黄体期卵泡颗粒细胞RNA,采用Illumina测序技术进行测序,并结合GO分析和KEGG通路分析对基因功能分类、参与的生物过程及信号网络进行了研究。试验共获得了17 627个可匹配基因的表达情况,发现了多个与繁殖、消化吸收及代谢相关基因的高表达,如CYP19A1、IL1A、IL1B、UCP2、STAR等基因。经过GO分析和KEGG通路分析,15 278个基因分别注释到三大类60个功能组,在biological process这一大类中发现了与繁殖、发育、代谢、调节和免疫等相关的功能组,有6 361个基因注释到了312个KEGG通路,发现了与生殖生物学、消化吸收、代谢等过程相关的信号通路,并对相关功能组和通路进行了分析。本试验揭示了湖羊黄体期卵泡颗粒细胞的基因表达特点及其参与的信号通路,为深入研究绵羊卵泡发育的调控机制及湖羊高繁性状关键基因的挖掘提供了参考。Follicular granulosa cells play an important role in the follicular development. This research investigated the gene expression and function of follicular granulosa cells during luteal phase in sheep. RNA were extracted from granulosa cells of Hu-sheep. Illumina expression profiling sequencing was performed on granulosa cells of Hu-sheep for the first time,particularly during luteal phase. GO analysis and KEGG pathway analysis were performed on detected genes. Totally 17,627 expressed genes were obtained. Some of the detected genes were associated to reproduction,digestion and absorption,metabolism. Many reproduction related genes were highly expressed in granulosa cells,such as CYP19A1,IL1 A,IL1B,UCP2,STAR,etc. Combining GO analysis and KEGG pathway analysis,15 278 of 17 627 detected genes were classified into three main categories,60 functional groups,some of them were related to reproduction,development,metabolism,regulation and immunity. There were 6361 genes annotated to 312 KEGG pathways,some of them were related to reproduction,digestion and absorption,metabolism. This study provided important genetic information resources for investigating the gene function,biological processes of granulosa cells of sheep.
