预针刺对应激性胃溃疡模型大鼠睡眠时间及CRH含量的影响

摘　　要：目的:观察预针刺对应激性胃溃疡模型大鼠的睡眠时间和血清、下丘脑中促肾上腺皮质激素释放激素(CRH)含量的影响。方法:32只雄性Wistar大鼠随机分空白组、模型组、和胃预针刺组、安神预针刺组,每组8只。和胃预针刺组选取双侧"足三里"及"中脘";安神预针刺组选取双侧"照海""申脉",针刺预处理10天后采用束缚-浸水(RWIS)应激法复制应激性胃溃疡模型。记录各组大鼠睡眠持续时间,放射免疫法检测血清、下丘脑中CRH的含量。结果:模型组大鼠的睡眠持续时间显著缩短,和胃组、安神组均延长睡眠持续时间,差异有统计学意义(P<0.01);模型组大鼠胃粘膜皱襞变平且颜色较暗,粘膜表面溃疡缺损形成,和胃组、安神组均可改善此病理变化,且睡眠的持续时间与胃溃疡指数呈负相关;模型组大鼠血清、下丘脑中CRH的含量与空白组比较差异显著升高,与模型组相比和胃组、安神组均有显著下降(P<0.01),组间无统计学差异。结论:胃粘膜损伤越严重,睡眠持续时间越短,针刺可有效地保护胃粘膜,预防应激性溃疡,延长造模后大鼠睡眠时间,其机制可能是通过调节中枢及外周CRH含量,进而调节神经内分泌系统,减轻机体的应激性反应。Objective： To investigate the effect on the expression of CRH in the serum and hypothalamus and the sleeping time with stress ulcer（ SU） by pre- acupuncture,to elucidate the possible role of pre- acupuncture in the development of SU and the sleep. Methods： 32 adult male Wistar rats were randomly divided into normal control group,model group,conciliatory stomach pretreatment group and sedative pretreatment group,and there were 8 rats in each group. We gave normal rats pre- acupuncture before modeling. For gastro- treatment group,acupoints at CV12 and ST 36 were chosen for needling and for dormancy group,acupoints at K6 and BL62 were used. After 10 days,use RWIS method to copy SU model rats. After that,the morphological changes of stomach mucosal tissue of those rats were observed. Meanwhile,the changes of sleep time which was induced by sodium pentobarbital were compared in each group. Results： The sleeping time of the model group was significantly lower than that in normal control group,while conciliatory stomach pretreatment group and sedative pretreatment group prolonged the time. In model group mucosal had large area of ulceration,bleeding,a large number of mucosal necrosis,shedding. While conciliatory stomach group and sedative group could reverse it. The expressions of CRH in the serum and hypothalamus in model group were increased significantly. Two kinds of pretreatments could reduce the content of CRH,and there was no statistical difference between two pretreatments.Conclusion： The worse of the gastric mucosa is,the less of the sleeping time is. Pre- acupuncture can protect the gastric mucosa effectively,enhance the rate of ulcer in rats and prolong rats＇ sleeping time. One of the mechanisms is probably by regulating the CRH content in the HPA axis and serum.
