摘  要:目的:探讨蒸发光散射检测器高效液相色谱法(HPLC-ELSD)测定桂红胶囊中银杏萜内酯含量的可行性。方法:色谱柱为Kromasil C18(250 mm×4.6 mm,5μm),流动相为甲醇-水(35∶65),流速1 m L/min,蒸发光散射检测器(ELSD)漂移管温度为105℃,载气流速3 L/min,柱温30℃。结果:白果内酯、银杏内酯A、银杏内酯B、银杏内酯C分别在2.04~20.40μg、2.08~20.80μg、1.36~13.60μg、1.22~12.20μg范围内呈线性关系,平均回收率分别为99.92%,98.10%,97.75%,99.31%,RSD值分别为2.25%,2.24%,1.17%,1.45%,回收率符合要求。结论:该方法准确可靠,专属性强,结果稳定,重现性好,可用于测定桂红胶囊中银杏萜内酯含量。Objective:To explore the feasibility of using high performance liquid chromatography evaporative light-scattering detector(HPLC-ELSD) to determine the content of ginkgo lactone in Guihong capsule. Methods:Kromasil C18 chromatographic column(250 mm × 4.6 mm,5 μm) was applied. The mobile phase consisted of methanol and water(35∶65) and the flow rate was 1 m L/min,the temperature of the ELSD drift tube was105 ℃,the air flow rate was 3 L/min and the column temperature was 30 ℃. Results:Bilobalide,ginkgolide A,ginkgolide B and ginkgolide C exhibit good linearity within the range of 2.04 μg to 20.40 μg,2.08 μg to20.80 μg,1.36 μg to 13.60 μg and 1.22 μg to 2.20 μg,respectively. Their average recovery rates were 99.92%,98.10%,97.75% and 99.31%,respectively,with RSD 2.25%,2.24%,1.17% and 1.45%,respectively. The recovery rate was qualified. Conclusion:This method was accurate,reliable,specific,repeatable,so it could be used for the determination of the content of ginkgo lactone in Guihong capsule.
