大黄水提物对雌性大鼠下丘脑、垂体结构的影响及可逆性研究

摘　　要：目的观察大黄水提物对雌性大鼠下丘脑、垂体结构影响及其可逆性,为临床安全用药提供实验依据。方法 SD雌性大鼠40只,随机取10只作为正常对照,其余30只灌胃大黄水提取物2.0g/kg(按生药计),连续给药60d。分别于末次给药后24h、停药30d、60d各处死总量的1/3。取脑、垂体,垂体称重、计算脏器指数;组织HE染色,光镜下观察组织形态学变化。结果给药60d垂体指数明显高于正常对照组(P<0.05);停药30d垂体指数与正常对照组比较差异无统计学意义(P>0.05)。停药30d垂体指数与给药60d比较差异无统计学意义(P>0.05);停药60d垂体指数明显低于给药60d(P<0.05)。组织形态学检测结果显示,给药组大鼠下丘脑神经元减少,有红色神经元,出现大量鬼影细胞及染色质边极现象;垂体细胞数量减少,细胞排列不规整,血窦严重扩张,嗜酸性细胞适应性增生,嗜碱性细胞减少。停药30d,下丘脑神经元有所增加,鬼影细胞及染色质边极现象减轻;垂体细胞数量增加,血窦扩张减轻,嗜酸性细胞适应性增生减少。停药60d,下丘脑神经元明显增加,鬼影细胞及染色质边极现象极少;垂体细胞数量明显增加,血窦扩张进一步减轻,嗜酸性细胞适应性增生明显减少。结论灌胃大黄60d可致雌性大鼠下丘脑、垂体组织结构损害,此种损害是可逆性的,停药60d基本恢复正常。Objective To observe the effect of Rhubarb water extract on the structure of hypothalamus and pituitary of female rats and reversibility,for providing the basis for clinical safety drug use.Methods Forty SD female rats,10 rats were randomly divided into normal control group,the other 30 were administration of Rhubarb water extract 2g kg-1for 60 days.After the 24 h of last administration,stopping the medicine for 30 days and 60 days,1/3 rats was respectively executed.Take the brain and pituitary,weighed and calculated the index of pituitary.The morphological changes of the tissues were observed by HE staining.Results The indexes of pituitary in medicine of 60 days were higher than that in normal control group obviously（P〈0.05）.The indexes of pituitary in stopping of 30 days compared with that in normal control group were no statistical significance（P〉0.05）.The indexes of pituitary in stopping of 30 days compared with that in medicine of 60 days were no statistical significance（P〉0.05）.The indexes of pituitary in stopping of 60 days were lower than that in medicine of 60 days（P〈0.05）.Pathological examination showed that the hypothalamus neurons reduce,red neurons,a lot of ghost cell and appearanced of chromatin edge extremely.Pituitary cell number decreased,blood sinus serious expansion,eosinophilic cell hyperplasia of adaptability,basophilic cells decreases.After stopping the medicine 30 days the hypothalamus neurons had increased,red neurons decreased,the phenomenon ghost cell and appearanced of chromatin edge extremely reduced.Pituitary cell number had increased,blood sinus serious expansion reduced,eosinophilic cell hyperplasia of adaptability reduced.After stopping the medicine 60 days,the hypothalamus neurons significantly increased,the phenomenon ghost cell and appearanced of chromatin edge very few.Pituitary cell number significantly increased,blood sinus serious expansion further reduced,eosinophilic cell hyperplasia of adaptability significantly reduced.Conclusion Gavage 60 days of
