利巴韦林抑制呼吸道合胞病毒诱导的A549细胞凋亡

摘　　要：目的:探讨利巴韦林抗呼吸道合胞病毒(RSV)的作用过程中,对A549细胞凋亡相关蛋白表达的影响。方法:将A549细胞分为4个组:细胞对照组(C组)、利巴韦林组(Rib组)、呼吸道合胞病毒组(RSV组)、利巴韦林干预组(RR组),分别在RSV感染的1,2,4,12,24h收集RSV组和RR组培养上清,空斑实验检测各个时间段病毒滴度;应用Western blot技术检测pAKT、AKT、IκBα、Bcl-2、Bax蛋白表达水平。结果:利巴韦林干预后,pAKT表达显著增加,差异有统计学意义(P<0.05);IκBα蛋白4,12,24h表达明显减少,差异有统计学意义(P<0.05);Bax蛋白表达明显减少,差异有统计学意义(P<0.05);Bcl-2蛋白12,24h表达高于RSV组,差异有统计学意义(P<0.05)。结论:利巴韦林不仅可以抑制RSV的复制,同时可以激活AKT-NF-κB途径、上调Bcl-2、下调Bax蛋白表达,从而抑制RSV感染诱导的A549细胞早期凋亡。Objective：To explore the role of protein expression related to apoptosis of A549 cells in the process of ribavirin against respiratory syncytial virus（RSV）.Methods：A549cells were divided into four groups：control group（group C）,ribavirin group（group Rib）,respiratory syncytial virus group（group RSV）and ribavirin treatment group（group RR）.By 1,2,4,12,24 hpost infection of RSV,culture supernatant of group RSV and group RR were collected,the viral titer of each time was tested with plaque assay.The expression of pAKT,AKT,IκBα,Bcl-2,Bax was detected by using Western-blot.Results：After the intervention,ribavirin pAKT expression increased significantly,the difference was statistically significant（P〈0.05）;IκBαpredominate protein 4h,12 h,24hexpression decreased significantly,the difference was statistically significant（P〈0.05）;Bax protein expression decreased significantly,the difference was statistically significant（P〈0.05）;The Bcl-2protein expression was higher than 12 h,24hRSV group,the difference was statistically significant（P〈0.05）.Conclusion：Ribavirin can not only inhibit replication of RSV,but also protect the host cell which is infected with respiratory syncytial virus by the activation of AKT-NF-κB pathway,up-regulating Bcl-2and down-regulating Bax expression.
