Plexin A1蛋白及mRNA在小鼠卵泡发育过程中的表达

摘　　要：目的:探讨Plexin A1在小鼠卵巢不同发育阶段的表达。方法:选择不同发育时期的C57BL/6j小鼠,采用免疫组织化学方法测定小鼠卵巢中Plexin A1蛋白水平的表达,实时定量聚合酶链反应(real-time PCR)方法检测Plexin A1mRNA水平的表达。结果:Plexin A1蛋白在小鼠卵巢各级卵泡的卵母细胞和颗粒细胞中均有表达。随着卵泡的不断发育,其在卵母细胞和颗粒细胞中的表达水平均逐渐增强。始基卵泡卵母细胞中的Plexin A1蛋白表达水平明显低于初级、次级和窦状卵泡的卵母细胞。始基卵泡颗粒细胞中未见明显Plexin A1蛋白表达,至初级卵泡阶段可见颗粒细胞开始有Plexin A1蛋白表达,其水平低于次级及窦状卵泡。闭锁卵泡中的Plexin A1呈强阳性表达,明显高于生长卵泡中的表达水平。Plexin A1mRNA在卵巢组织中的表达水平随周(日)龄呈先升高后降低的趋势,5d,7d,3周小鼠卵巢中Plexin A1 mRNA表达水平分别为3d小鼠的1.56、1.26、0.79倍(P<0.05),而2周小鼠卵巢中的Plexin A1mRNA表达水平与3d小鼠相当(P>0.05)。结论:Plexin A1在不同级别的小鼠卵泡中均有表达,且表达水平随卵泡的不断生长发育而增强,但闭锁卵泡中表达最强,提示Plexin A1在卵泡的生长发育及闭锁过程中可能发挥重要的调控作用。Objective：To investigate the spatial and temporal expression pattern of protein and mRNA of Plexin A1 in mouse ovary.Methods：Mouse ovaries of different development stage were collected.The expressions of Plexin A1 protein were detected by immunohistochemistry,and the mRNA expressions were detected and evaluated by real-time PCR.Results：Plexin A1 protein was expressed both in oocyte and granulosal cells of different development stages.The expression of Plexin A1 protein increased in accord with the follicular development.Plexin A1 protein level in oocyte of primordial follicles was significantly lower than that in primary,secondary and antral follicles.The protein level in granulosal cells of primordial follicles could be hardly found.Granulosal cells of the primary follicles started to express Plexin A1 protein,but the expression levelwas much lower than those of secondary and antral follicles respectively.The expression level of Plexin A1 protein in the atretic follicles was much higher than those of the growing follicles.The Plexin A1 mRNA expression level in ovary tissues increased at first and then decreased in accord with the age before 3weeks old.The expression level of Plexin A1 mRNA in the ovary of 5days old,7days old and 3weeks old were 1.56,1.26,0.79 times of those in 3days old（P〈0.05）.The expression level of 5days old was the same as those in 3days old（P〈0.05）.Conclusion：Plexin A1 protein and mRNA express in the mouse ovary,especially at the late stage of follicular development and atretic follicles,which suggests that Plexin A1 may be involved in folliculogenesis and atresia.
