摘 要:目的:研究P物质(SP)在慢性应激引起的大鼠结肠动力紊乱中的作用及其机制。方法:采用SPF级Wistar大鼠,体重180-200g,随机分为模型组(WAS组,即避水应激组)和对照组(SWAS组,即假避水应激组)。WAS组采用经典避水应激模型,每天给予避水应激处理1h,连续10d。造模结束后,用10%水合氯醛麻醉大鼠,分别留取心脏采血标本及结肠肌层标本。采用酶联免疫分析法检测血清SP水平,采用实时荧光定量PCR技术检测结肠肌层神经激肽受体1(NK1R)mRNA的表达水平。此外,应用膜片钳技术检测结肠平滑肌细胞L型钙电流的变化。结果:WAS组大鼠血清SP水平显著高于SWAS组(P<0.01),WAS组结肠肌层NK1R mRNA表达水平明显高于SWAS组(P<0.05),WAS组大鼠平滑肌细胞的峰电流加SP前后的比值与SWAS组比值相比,差异有统计学意义(P<0.05)。结论:慢性应激可诱导结肠肌层NK1R表达增加,而SP可通过作用于表达于结肠肌层NK1R引起结肠动力的改变,在肠易激综合征的肠动力紊乱中发挥重要作用。Objective:To study the effect and mechanisms of substance P(SP)on colonic smooth muscle contractility disorders induced by chronic water avoidance stress(WAS).Methods:Wistar rats were divided into two groups randomly,including WAS group and SWAS group.The WAS group was given the typical water avoidance stress model,suffering from WAS for one hour daily for 10 consecutive days.On the last day,the rats were anesthetized by 10% chloral hydrate,then the heart blood and colonic smooth muscle samples were taken.The concentration of SP was measured through ELISA,and the expression of NK1 R mRNA on the colonic smooth muscle was measured by the real-time quantitative PCR.Furthermore,the patch clamp recording technique was applied to measure the L-type calcium current of the colonic smooth muscle.Results:The concentration of SP in WAS group was significantly higher than that in SWAS group,and the expression of NK1 R mRNA of colonic smooth muscle in WAS group was also significantly higherthan that inSWAS group,and the peak current ratio of before and after adding SP in WAS group was significantly different from SWAS group,and the difference between the two groups had statistical significance.Conclusion:SP may be involved in the colonic smooth muscle contractility disorders induced by chronic water avoidance stress through the NK1 Rexpression in the colonic smooth muscle.
