摘  要:【目的】开展贵州地区不同山羊品种的黑皮质素受体-3(MC3R)基因转录水平研究,为研究山羊MC3R的作用部位、特点及功能提供理论参考。【方法】以黔东南小香羊、贵州白山羊、贵州黑山羊、黔北麻羊和南江黄羊5个山羊品种为研究对象,利用双标准曲线法和Taq Man实时荧光定量PCR对肝脏、肾脏、心脏、肺脏、背最长肌、半膜肌和皮下脂肪中MC3R基因的转录水平进行检测。【结果】MC3R基因在山羊的心脏、肝脏、肺脏、肾脏、背最长肌、半膜肌、皮下脂肪中均转录成m RNA,其中在皮下脂肪、肾脏、肝脏高度转录,在肺脏中度转录,而在心脏、背最长肌、半膜肌中的转录水平较低。7个组织中,MC3R基因转录水平均以贵州白山羊最高。【结论】山羊MC3R基因的转录水平存在组织及品种间差异,在非肌肉类组织中的转录水平高于在肌肉类组织,且在贵州白山羊组织中转录水平高于其他品种,在今后研究MC3R基因与生长和肉质性状关联性时应注意取材品种和部位对其转录水平的影响。【Objective】The transcriptional level of melanocortin receptor 3(MC3R) in different goat breeds were investigated, in order to provide theoretical references for researching action sites, characteristics and functions of MC3 R in goat. 【Method 】Taking Qiandongnan small-Xiang goat, Guizhou white goat, Guizhou black goat, Qianbei Ma goat and Nanjiang yellow goat as research objects, the double standard curve method and real-time fluorescence quantitative PCR technique was used to determine MC3 R gene transcriptional levels in liver, kidney, heart, lung, longissimus muscle,semimembranosus muscle and subcutaneous fat. 【Result】The results showed that MC3 R gene was transcribed into m RNA in heart, liver, lung, kidney, longissimus dorsi, semimembranosus and subcutaneous fat of goat. However, MC3 R gene was highly transcribed in subcutaneous fat, kidney and liver, moderately transcribed in lung, and lowly transcribed in heart,longissimus dorsi and semimembranosus. Among five goat breeds, Guizhou white goat had the highest MC3 R gene transcriptional level in 7 tissues. 【Conclusion】There exist differences in MC3 R gene transcriptional level between breeds and between tissues. The transcriptional level in non-muscle tissues are higher than that in muscle tissues. Moreover, Guizhou white goat are higher transcriptional level than other goat breeds. Therefore, it is suggested that effects of sampling breeds and tissues on MC3 R gene transcriptional levels should be paid more attention to when studying correlation between MC3 R gene and growth and meat quality traits.
