糖尿病肾病大鼠中医气虚模型的制作及其影响因素

摘　　要：目的探讨糖尿病肾病(DN)大鼠气虚模型的制作方法和成模影响因素。方法将43只雄性SPF级大鼠随机分为正常组(n=7)、实验组(n=36)。正常组提供正常饮食,实验组以高脂饲料饲养联合链脲佐菌素(分别予一次大剂量注射、多次小剂量注射)制模成功后,随机分为DN组和DN气虚组,DN气虚组予高脂连续喂养+疲劳游泳试验造模,监测造模后大鼠的体质量、尿量、血糖、尿微量白蛋白及肾脏病理的改变情况。结果实验组DN造模成功率为83.3%(30/36),其中一次大剂量注射法成功率为77.8%,多次小剂量注射法成功率为88.9%。DN气虚组中,游泳劳损法成功率为60%(9/15),死亡率为26.1%(6/23)。随造模时间的延长,DN组及DN气虚组的体重降低,24 h尿微蛋白增加,而尿量、血糖、24 h尿蛋白定量则呈现先升高后回落的趋势。结论DN大鼠气虚模型制备成模率较高,但受多因素影响。Objective To explore the method of establishing the rat model of Qi- deficiency and diabetic nephropathy and its influential factors. Methods 43 SPF male rats were divided randomly into 2 groups,the normal group（ 7 rats） and the model group（ 36 rats）. The normal group was treated with normal diets. The model group were fed with high- fat diet followed by injection of STZ and were further divided randomly into the DN model group and DN with Qi-deficiency group. Rats in the DN with Qi- deficiency group were continuously fed with high- fat diet and underwent fatigue swimming test. Weight,urine volume,blood glucose,urinary microalbumine and pathologic changes in the kidney were monitored. Results The successful rate of STZ injection in establishment of the rat model of diabetic nephropathy was 83. 3%,with 77. 8% for single large- dose injection and 88. 9% for multiple injections. The successful rate and mortality of DN with Qi- deficiency group were 60% and 28. 6%,respectively. With increased duration of treatment,weight increased and 24 h urinary microalbumine decreased,while urinary volume,blood glucose and 24 h urinary protein decreased after initial rising. Conclusion The successful rate of the rat model of Qi- deficiency and diabetic nephropathy is high,but affected by many factors.
