摘 要:[目的] 对外观相近、较难区分的4个白杨新品种及其父母本和3个近缘种进行遗传分析并构建指纹图谱,为这些品种在生产应用中的分子鉴定提供技术依据。[方法] 采用SRAP分子标记技术,对03-4-9、03-4-22、03-5-17和03-6-11 4个白杨新品种及I-101杨、截叶毛白杨、84K、毛白杨30号、银毛杨共9个白杨品种开展遗传分析及指纹图谱构建。[结果] 14对多态性高的SRAP引物对9个白杨品种共扩增出167条条带,平均每对引物扩增条带为11.93条。其中多态性条带143条,多态性比率为85.63%,多态性高。聚类分析表明,9个白杨品种之间的遗传相似系数为0.40~0.76,其中4个白杨新品种与父本截叶毛白杨和毛白杨30号亲缘关系较近,遗传相似系数平均值分别为0.65和0.63,与母本I-101亲缘关系稍远,遗传相似系数平均值为0.49。利用重复性好的3对引物(me5/em10、me7/em2和me7/em3)扩增图谱构建了9个白杨品种的指纹图谱,每对引物均可将9个白杨品种单独区分开。[结论] 9个白杨品种在SRAP位点有较高的多态性,SRAP分子标记能很好地用于杨树品种的鉴定及指纹图谱构建。[Objective]Genetic relationship and DNA-fingerprinting of 9 Populus L.in Sect.Leuce Duby verities(4new hybrids,male and female parents,and 3related verities)that were difficult to be distinguished by appearance were investigated using molecular markers technique to improve molecular identification during production.[Method]SRAP molecular markers were used to conduct genetic analysis and build fingerprints for 03-4-9,03-4-22,03-5-17,03-6-11,P.alba(Italy),P.tomentosavar.truncata,P.alba×P.glandoulsa,P.tomentosa No.30,and P.alba(China)×P.tomentosa.[Result]A total of 167 bands were amplified with 14 pairs of primers,with the mean of 11.93 per primer.There were 143 polymorphic bands with a ratio of 85.63%.Cluster analysis showed that the genetic similarity coefficients among the 9varieties were 0.40-0.76,and 4new hybrids had close genetic relationship with P.tomentosa var.truncata(male parent)and P.tomentosa No.30 with the mean coefficients of 0.65 and 0.63,respectively.The genetic relationship between 4new hybrids and P.alba(Italy,female parent)was less close with the meancoefficient of 0.49.The fingerprints of 9varieties were established by amplification map of 3repeatable primers(me5/em10,me7/em2,and me7/em3),and the 9varieties could be identified separately.[Conclusion]The 9poplar varieties had high polymorphism in SRAP sites,and SRAP molecular markers technique could be used for identification and fingerprint establishment of Poplar varieties.
