摘  要:目的评价金黄色葡萄球菌(SAU)、凝固酶阴性葡萄球菌(CNS)的两种细菌生物膜表型检测法,探讨生物膜表型特点与icaA/icaD基因的关系。方法 2007年8月-2014年12月共收集90株SAU和168株CNS,采用刚果红琼脂平皿和半定量生物膜检测法确定生物膜表型,PCR法检测icaA/icaD基因。结果刚果红试验的168株CNS中生物膜阳性32株,阳性率为19.0%;90株SAU中生物膜阳性32株,阳性率为35.6%;半定量检测的CNS中生物膜阳性38株;SAU中生物膜阳性36株;两种检测方法比较差异无统计学意义;PCR检测结果表明icaA/icaD基因两者同时含有或同时缺乏,尚未发现单一携带者,红色变异体的icaA/icaD基因阴性。结论可采用更简便、更直观的刚果红法筛查生物膜,icaA、icaD基因是生物膜产生的基础。OBJECTIVE To evaluate two biofilm phenotypic tests for Staphylococcus aureus(SAU)and coagulase negative Staphylococcus(CNS),and to investigate the relationship between biofilm phenotypic characteristics and icaA /icaD genes.METHODS 168 CNS and 90 SAU were collected from Aug.2007 to Dec.2014.Congo red technique and semi-quantitative biofilm assays were performed to determine biomembrane type.The icaA /icaD gene were determined by the polymerase chain reaction method.RESULTS With Congo red test,32 of 168CNS(19.0%)and 32 of 90SAU(35.6%)were found to be positive.The semi-quantitative biofilm assays indicated that 38CNS(22.6%)and 36SAU(40.0%)were positive.There was no significance between two phenotypic detection methods.PCR method suggested that the strains either carried both of icaA and icaD or lacked of both.Single carriers had not been found.The icaA /icaD were negative in red variants.CONCLUSION More convenient and intuitive Congo red method can be used to screen biofilm.IcaAand icaD genes are the basis of the biofilm production.
