炭样小单孢菌JXNU-1产抗生素JX对嗜根考克氏菌肽聚糖合成的影响及其机理

摘　　要：为了揭示一株具有广谱抗菌活性炭样小单孢菌JXNU-1产的核苷类抗生素JX对嗜根考克氏菌肽聚糖合成的影响,本研究采用超声破壁、称重法研究抗生素对嗜根考克氏菌细胞壁含量变化,采用iTRAQ技术对抗生素处理前、后嗜根考克氏菌的蛋白质组进行比较分析。结果显示,在抗生素JX作用下,嗜根考克氏菌细胞分裂被抑制,细胞壁含量升高;iTRAQ技术鉴定了抗生素胁迫下的嗜根考克氏菌细胞中的1 780个蛋白,其中差异表达蛋白149个,包括表达上调蛋白106个,表达下调蛋白43个,上调表达蛋白中包括有一与肽聚糖合成的相关酶MurG。本研究为揭示炭样小单孢菌JXNU-1产抗生素JX的抗菌作用机制提供了基础数据。In order to reveal the effects and its mechanism of the nucleoside antibiotics,termed the antibiotics JX from a strain of Micromonospora carbonacea JXNU-1 with broad-spectrum antimicrobial activity,on the biosynthesis of cell peptidoglycan in Kocuria rhizophila,the change of cell wall content in Kocuria rhizophila was studied with Cell wall weighing method after ultrasonic cell disruption,and the comparative proteomics of Kocuria rhizophila was analyzed,with isobaric tags for relative and absolute quantization（iTRAQ）. The results showed that the cell division of K. rhizophila was inhibited,and the content of cell wall of K. rhizophila was increased under the influence of antibiotics JX. Based on i TRAQ,1 780 proteins were identified in the cell of K. rhizophila under the stress of antibiotics. Among them,149 were differential expression protein,including 106 proteins up-regulated and 43 proteins down-regulated. The up-regulated proteins contained the enzyme Mur G which involved in the biosynthesis of peptidoglycan. This research provided the basic data about antimicrobial mechanisms of the antibiotics JX from M. carbonacea JXNU-1.
