摘  要:为奶牛性控精子体外受精提供具有活力的精子,筛选出高效的精子分离方法来提高体外受精效率。将市售的冷冻奶牛性控精液解冻、精子浓度调节为1×107cells/m L后,利用微流路精子分离器(microfluidic sperm sorter,MFSS)分离精子,检测分离后的精液质量并分析精子的线粒体膜电位变化。精液样本通过MFSS分离后,精子活力由35.28%提高到57.24%(P<0.05);直线速度由23.91μm/s提高到32.76μm/s(P<0.05)。此外,MFSS的C室中精子线粒体相对活性为1.56,显著高于分离前A室(0.28)的(P<0.05)。综上结果 ,微流路精子分离器可以分离出既有较高活力又具有高活性的线粒体的奶牛性控精子,精子质量得到较大的改善。This research aimed at isolating high motility sperm for in vitro fertilization, and screening out the highly effective selection method of viable sperm. The commercially frozen sex-sorted semen of dairy cow was thawed and the sperm concentration was adjusted to 1 ×10^7cells/m L, quality of sperm before and after dealing with selection technique of MFSS(Microfluidic sperm sorter, MFSS) and swim-up methods were analyzed by CASA. Meanwhile, mitochondrial membrane potential changes of the sperm was analyzed by Flow cytometry The results showed that emen samples before and after separation by MFSS, sperm quality had obviously been improved, and sperm motility was increased from 35.28% to 57.24%(P〈0.05), and Linear speed was increased from 23.91 μm/s to 32.76 μm/s(P〈0.05). The results showed that the sex-sorted sperm with high activity and high activity of mitochondria were isolated by MFSS,and quality of spermatozoa could be greatly improved.
