摘 要:目的构建猪小肠黏膜下层脱细胞组织基质(SIS)材料,并与美国COOK公司的Surgisis产品进行对比,评估SIS材料的生物相容性检验。方法按照文献中报道的方法制作SIS材料,并与美国COOK公司的Surgisis产品进行大鼠体内植入的对比,观察并记录动物术后的一般情况,并于术后3、7 d、2、4、8周处死大鼠(每次2只),分别切取SIS及Surgisis补片并包含周围部分正常的肌肉筋膜组织,观察材料-肌肉交界处局部炎症反应、组织增生及微血管形成的情况。结果所有动物均存活至既定处死时间点,组织学观察可见,术后早期,SIS及Surgisis结构完整,呈现膜片状或条索状致密红染结构,植入部位可见大量炎性细胞浸润;随着时间的延长,SIS及Surgisis逐渐降解,结构松散,炎性细胞逐渐减少。结论 SIS植入大鼠体内后具有良好的生物相容性,未出现明显的排斥反应。Objective To construct the small intestinal mucosa,and to evaluate the in vivo biocompatibility of SIS compared with the American COOK company' s product-Surgisis. Methods We constructed the SIS material by using the method reported in the literature, and tested the in vivo biocompatibility of SIS with the American COOK company' s product-Surgisis via Rat model. General conditions of the animals were observed and recorded postoperatively,such as wound infection,sinus or fistula formation,death and so on. At 3 days,7 days,2 weeks,4 weeks and 8 weeks after implantation,two rats were sacrificed every time; and the specimen,including the biomaterials and surrounding muscle fascia tissue,was obtained and to detect the infiltration of inflammatory cells with haematoxylin and eosin( HE)stain. Results All animals survived to the established time point with no accidental death occurred. The SIS and Surgisis both showed an acute inflammatory response by extensive inflammatory cell infiltration in histology at the implanted site at 3 and 7 days after implantation. SIS and Surgisis both kept the integral structure,which showed membrane or cord-like dense red structure under optical microscope with HE stain.Degradations of both SIS and Surgisis and reduction of inflammatory cell infiltration were observed at 4 and 8weeks after implantation. Conclusion The result shows that there was good in vivo biocompatibility of SIS after planted in animals without severe rejection.
