摘  要:目的采用HPLC法测定肾移植患者血浆中的霉酚酸酯活性代谢物霉酚酸。方法采用Symmetry C18柱(150 mm×3.9 mm,5μm),流动相为乙腈-32 mmol·L^-1甘氨酸缓冲液(p H9.2)(17∶83),柱温30℃,流速1.0 m L·min^-1,用荧光检测法,激发波长342 nm,发射波长425 nm,进样量为20μL。结果霉酚酸0.01~40.00μg·m L^-1与峰高的线性关系良好(r=0.999),最低定量限为0.01μg·m L^-1;提取回收率、加样回收率分别为92.29%~95.01%、100.69%~110.57%,日内、日间RSD均〈8%。结论所用方法灵敏、快速、准确,符合生物样品分析方法的基本要求,适用于霉酚酸血药浓度的测定。OBJECTIVE To establish an HPLC method for the determination of mycophenolic acid( MPA) in the patients plasma of kidney transplant. METHODS The separation was performed on a Symmetry C18( 150 mm × 3. 9 mm,5 μm) with a mobile phase of acetonitrile- 32 mmol·L^-1glycine- Na OH buffer solution( p H9. 2)( 17 ∶83) at a flow rate of 1. 0 m L·min^-1,and the fluorescence detector of 342 /425 nm. RESULTS A good linearity was obtained over the range of 0. 01- 40. 0 μg·m L^-1( r = 0. 999). The extraction recovery and method recovery were 92. 29%- 95. 01% and 100. 69%- 110. 57%. The inter- day and intra- day RSD were all less than 8%. CONCLUSION This method is rapid,sensitive and specific for the determination of mycophenolic acid in the patients plasma of kidney transplant in clinical pharmacokinetic and mycophenolic acid drug monitoring study.
