摘 要:通过比较5种不同的播前预处理方法,探究蒙古黄芪种子在自然条件下萌发率较低的原因,并确定提高黄芪种子萌发率的最优方案。结果表明,与对照组比较,砂磨、80%浓硫酸、沸水浸种与环割处理均能够显著提高黄芪种子萌发率,而98%硫酸处理却降低了黄芪种子的萌发率;用80%浓硫酸处理、温水浸泡和400 mg/L GA_3处理,黄芪种子萌发率显著高于单纯的80%浓硫酸处理。该结果说明黄芪种子种皮过厚和内源性抑制物共同限制了其种子的萌发;用80%浓硫酸处理2 min后,在40-45℃温水中浸泡24 h,再用400 mg/L的GA_3处理24 h,为生产实践中提高黄芪种子萌发率的最优方案。Five pre-sowing methods were compared to find out the main causes that inhibiting the seed germination of Astrgalus menbranceus, and determine the optimal method to enhance the germination rate of A.menbranceus. The seeds were treated by five different ways before sowing. The results showed that seed coat and the endogenous inhibitors were main causes which limited the seed germination. Treating seeds in 80% H_2SO_4, sanding or ringing, soaking in water could significantly increase the seed germination rate, while soaking in 98% H_2SO_4 could significantly decrease the seed germination rate; the seed germination under the treatments of soaking in 80% H_2SO_4, washing in warm water for 24 h plus 400 mg/L of GA_3 was much higher than individual treatment of 80% H_2SO_4. The results suggested that both seed coat and endogenous inhibitors limit the seed germination of A.Menbranceus, and the optimal method of increasing the germination rate of A.menbranceus was soaking seeds in 80% H_2SO_4 for 2min, and washing in 41-45 ℃ water for 24 h, then treating with 400 mg/L of GA_3 for 24 h.
