水解酪蛋白粉增强中药复方降血压作用的研究

摘　　要：目的:考察水解酪蛋白粉(Casein hydrolysate,CH)对天麻、钩藤和杜仲复方(Chines herbal compound,CHC)降血压作用的影响和急性毒性。方法:将SD大鼠分为空白对照组和样品正常组,给予蒸馏水和CH与CHC配伍药物;将原发性高血压大鼠(SHR)分为牛黄降压胶囊组、CH-CHC药物低、中、高剂量组、CH组和CHC组及模型对照组,每天灌胃给药1次,连续4周,记录大鼠收缩压、舒张压、心率和血清一氧化氮(NO)、血管紧张素Ⅱ(AngⅡ)、内皮素-1(ET-1)含量;急性毒性实验以KM小鼠按最大给药体积灌胃,每天2次,连续14天,记录动物一般变化和主要脏器系数。结果:与模型对照组比较,CH-CHC中高剂量组、CH组和CHC组大鼠收缩压和舒张压显著降低(P<0.05或P<0.01),CH-CHC降血压作用显著优于CH组和CHC组(P<0.05),相应血清AngⅡ、NO和ET-1水平也具有显著性差异,各药物组对大鼠心率均无显著影响,急性毒性试验药物组与对照组结果无显著差异。结论:CH可以显著增强天麻、钩藤和杜仲复方的降血压作用,两者配伍无急性毒性作用。Objective： To investigate the effect and acute toxicity of Casein hydrolysate（CH） on Chinese herbal compound（CHC） of gastrodia elata,uncaria and eucommia in reducing blood pressure. Methods：SD rats were divided into the control group and CH- CHC normal group, CH and CHC compatibe drugs and distilled water being given. Rats with essential hypertension were divided into niuhuang jiangya capsules group,CH- CHC drugs low,mid and high dose groups,CH group,CHC group,oral administration once every day for 4 weeks. Recorded systolic pressure,diastolic pressure,heart rate and contents of serum nitric oxide（NO）,angiotonin Ⅱ（AngⅡ）,endothelin- 1（ET- 1）. KM rats were gavaged according the maximum dose volume on the acute toxicity test,2 times every day for 14 days. Recorded the common changes and the main organ coefficient of animal. Results：Comparing with those in the model group,the systolic pressure and diastolic pressure of the rats in CH- CHC mid and high dose group,CH group and CHC group were dropped significantly（P〈0.05 or P〈0.01）,the effect of CH- CHC reducing blood pressure being obviously superior to that in CH group and CHC group（P〈0.05）,so as the levels of corresponding serum AngⅡ,NO and ET- 1. There was no significant effect on heart rate of rat in every drug group,and there was also no difference between the acute toxicity drug group and the control group. Conclusion：CH can improve the effect of CHC of gastrodia elata,uncaria and eucommia in reducing blood pressure. The compatibility of the two drugs is no acute toxicity.
