摘  要:目的:观察丹蒌片对动脉粥样硬化(atherosclerosis,AS)模型大鼠血清抵抗素(Resistin)和血管内皮的影响,探讨丹蒌片对AS模型大鼠血管内皮的保护作用及可能机制。方法:32只雄性Wistar大鼠随机分为正常组、模型组、他汀组、丹蒌组,每组8只。正常组大鼠喂食基础饲料12周,余各组均采用高糖高脂饮食+维生素D3腹腔注射方法复制AS模型。模型组、他汀组、丹蒌组,分别给予生理盐水、阿托伐他汀片混悬液、丹蒌片混悬液灌胃干预8周。干预结束后,行ELISA法检测大鼠血清Resistin、内皮素-1(ET-1)、白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和一氧化氮(NO)的水平。结果:1与正常组比较,模型组大鼠血清Resistin、ET-1、IL-6和TNF-α水平升高(P<0.05,P<0.01,P<0.05,P<0.05),NO水平下降(P<0.05);2与模型组比较,他汀组大鼠血清Resistin、ET-1、IL-6和TNF-α水平降低,差异有统计学意义(P<0.01,P<0.01,P<0.01,P<0.05),NO水平上升(P<0.01);丹蒌组大鼠血清Resistin、ET-1、IL-6和TNF-a水平降低,差异有统计学意义(P<0.01,P<0.01,P<0.01,P<0.01),NO水平上升(P<0.01);3他汀组与丹蒌组之间大鼠血清resistin、ET-1、IL-6、TNF-α和NO之间水平差异无统计学意义(P>0.05)。结论:1丹蒌片可降低AS模型大鼠血清Resistin、ET-1、IL-6和TNF-α水平,升高NO水平,减轻AS模型大鼠血管内皮的损伤,作用与他汀类药物相似。2丹蒌片可保护血管内皮,其作用机制可能是通过降低血清Resistin、ET-1、IL-6和TNF-α水平,增加NO的分泌,从而保护血管内皮细胞结构的完整和功能的完善。Objective:To observe the effect of Danlou tablets on serum resistin and vascular endothelial injury in the rat model of atherosclerosis(AS) and discuss the protective effect and possible mechanism of Danlou tablets in the rat model of AS. Methods:Thirty- two of male Wistar rats were divided into the normal group,the model group,statin group and Danlou group randomly,8 cases in each group. The rats in the normal group were fed by basal feed for 12 weeks;the rats in other groups were fed by high- sucrose- fat diet and vitamin D3 intraperitoneal injection to copy AS models. The rats in the model group, statin group and Danlou group were given respectively normal saline, suspension of atorvaststin, suspension of Danlou tablets for lavage intervention for 8 weeks. After intervention,detected the levels of rat serum resistin,endothelin-1(ET- 1),interleukin- 6(IL- 6),tumor necrosis factor- α(TNF- α) and nitric oxide(NO). Results:Comparing with those in the normal group,the levels of serum resistin,ET- 1,IL- 6 and TNF- α in the model group were all increased(P〈0.05,P〈0.01,P〈0.05,P〈0.05),the level of NO were dropped(P〈0.05). Comparing with those in the model group,the levels of serum resistin,ET- 1,IL- 6 and TNF- α in the statin group were all dropped(P〈0.01,P〈0.01,P〈0.01,P〈0.05),the levels of NO were increased(P〈0.01);the levels of serum Resistin,ET- 1,IL- 6 and TNF- α in the Danlou group were all dropped(P〈0.01,P〈0.01,P〈0.01,P〈0.01),the levels of NO were increased(P〈0.01). The difference of serum resistin,ET- 1,IL- 6, TNF- α and NO between the statin group and Danlou group were not significant(P〉0.05). Conclusion: Danlou tablets can reduce the levels of serum resistin,ET- 1,IL- 6 and TNF- α,and improve the levels of NO,and relieve vascular endothelial injury in AS model rats.
