摘 要:目的:探讨不同方式运动对自然衰老小鼠骨骼肌自噬的影响及其分子生物学机制。方法:36只8周龄雌性小鼠随机分为衰老对照组(C)、衰老自主运动组(V)、衰老跑台运动组(F)。C组于鼠笼中正常饲养,V组和F组分别进行8周的自主跑轮运动和跑台运动。训练结束12~24h内,取腓肠肌,利用RT-PCR法检测相关细胞因子的mRNA表达。结果:与C组相比,V组PI3K(P〈0.05)、PDK1(P〈0.05)和Atg3(P〈0.05)的mRNA表达均出现显著变化;与C组相比,F组PI3K(P〈0.01)、PDK1(P〈0.01)、Akt(P〈0.05)、m TOR(P〈0.05)、TSC2(P〈0.05)、Atg3(P〈0.01)、ULK1(P〈0.05)、Beclin1(P〈0.05)、LC3(P〈0.05)和Atg7(P〈0.05)mRNA表达均出现显著变化;与V组相比,F组PI3K(P〈0.05)、PDK1(P〈0.05)、Akt(P〈0.05)的mRNA表达均显著上调。结论:跑台运动可通过激活PI3K/Akt/m TOR信号通路抑制衰老小鼠骨骼肌的自噬水平。Objective: Research the influence of different exercise modes on autophagy of muscles in aging female mice and the biology mechanism. Methods: 36 8-week old female C57 BL /6 mice were randomly divided into three groups: aging control group( C),aging voluntary wheel running group( V) and aging treadmill running group( F). After one week,group V and group F respectively did voluntary wheel training and treadmill training. After 12 h of the last training,using the mothod of RT-PCR to detect the mRNA expression of related gene in the gastrocnemius muscles. Results: Compared to group C,the mRNA expression of PI3K( P〈0. 05),PDK1( P〈0. 05) and Atg3( P〈0. 05) of group V were significantly up-regulated. Compared to group C,the mRNA expression of PI3K( P〈0. 01),PDK1( P〈0. 01),Akt( P〈0.05),m TOR1( P〈0. 05),TSC2( P〈0. 05) 、Atg13( P〈0. 05) and ULK1( P〈0. 05) of group F were significantly upregulated and the expression of mRNA of Beclin1( P〈0. 05) and Atg7( P〈0. 05) significantly down-regulated. Compared to group C,the expression of mRNA of PI3K( P〈0. 01),PDK1( P〈0. 01),Akt( P〈0. 05),m TOR1( P〈0.05),ATG13( P〈0. 01),ULK1( P〈0. 05),Beclin1( P〈0. 05),LC3( P〈0. 05) and Atg7( P〈0. 05) were all significantly changed. Compared to group V,the mRNA expression of PI3K( P〈0. 05) 、PDK1( P〈0. 05) and Akt( P〈0. 05) were all significantly up-regulated. Conclusion: Treadmill training can restrain the autophagy level of the aging mice muscles through activating the PI3 K / Akt / m TOR signaling pathway.
