摘 要:目的研究纳米PEEK/HA复合材料对MG63细胞相容性的影响。方法应用流式细胞仪技术研究不同组分的复合材料浸提液中MG63细胞增殖的情况;应用实时定量RT-PCR技术检测复合材料浸提液中MG63细胞特征性蛋白碱性磷酸酶和Ⅰ型胶原的表达情况。结果流式细胞仪实验结果显示,4种浸提液中培养的MG63细胞,48 h后,均有稳定的增殖。10%HA/SPEEK/PEEK组增殖指数为59.3%,高于其他4组,差异有统计学意义。实时定量RT-PCR实验结果显示,4种材料的浸提液培养MG63细胞于14、21、28 d时成骨细胞特征性的蛋白ALP、COLI的DNA的量表达明显。10%nHA/SPEEK/PEEK组表达高于其他4组,差异有统计学意义。随着时间的延长,各组的表达均有所下降。结论纳米PEEK/HA复合材料对MG63细胞有良好的生物相容性。Objective To study the effect of PEEK/HA composite materials on MG63 cell compatibility. Methods Flow cytometry (FCM) was used to study proliferation of MG63 in different components of the composite material leaching liquors. Real-time quantitative RT-PCR was adopted to detect the expression of proteins characteristic of alkaline phosphatase and type Ⅰ collagen of MG63 cells in different components of the composite material leaching liquors. Results Flow cytometry results showed that MG63 cells cultured in the 4 leaching liquors at 48 hours had stable growth. The proliferative index in the 10% HA/SPEEK/PEEK group was 58.7% which was higher than that in the other three groups and the control group. The difference was statistically significant. Real-time quantitative RT-PCR results showed in MG63 cells cultured by 4 leaching liquors at 14 days, 21 days, 28 days, the expression of the characteristic protein ALP, and COLI of osteoblasts was obvious, especially in the 10% nHA/SPEEK/PEEK group which was higher than that in the other three groups and control group, the difference was significant. As time passed, the expression in each group has declined. Conclusion PEEK/HA composite materials have good biocompatibility with MG63 cells.
