摘 要:[目的]探讨枳实总黄酮苷提取物(aurantii fructus immaturus flavonoid,AFIF)对功能性消化不良(functional dyspepsia,FD)模型大鼠胃适应性的影响及其可能的作用途径。[方法]10日龄雄性SD大鼠86只,随机分为正常组22只及造模组64只,造模组采用碘乙酰胺(iodoacetamide,IA)灌胃法联合夹尾应激法诱导大鼠FD模型;至大鼠8周龄时将造模组大鼠随机分为枳实总黄酮苷提取物大、中、小剂量组及模型组,分别予大、中、小剂量的枳实总黄酮苷提取物或生理盐水干预14d,给药结束后,各组大鼠经食道置入胃内气囊,应用电子恒压器对胃内气囊分阶段加压,检测大鼠胃适应性,加压程序为:以1mmHg为起始压力,维持10min后,每分钟压力升高1mmHg直至10mmHg(斜坡期),在10mmHg压力下维持10min(平台期)。应用离体平滑肌张力测定技术检测不同浓度的枳实总黄酮苷提取物对大鼠近端胃体离体纵行平滑肌张力的影响。[结果]近端胃适应性检测结果显示:在斜坡期及平台期,模型组大鼠胃适应性较正常组显著降低(P<0.05);枳实总黄酮苷提取物大剂量组(大剂量组)大鼠胃适应性在斜坡期及平台期均较模型组显著升高(P<0.05);枳实总黄酮苷提取物中剂量组(中剂量组)大鼠胃适应性在平台期较模型组显著升高(P<0.05);枳实总黄酮苷提取物小剂量组(小剂量组)大鼠胃适应性较模型组差异无统计学意义。近端胃体离体平滑肌张力测定结果显示:近端胃体纵行平滑肌张力随枳实总黄酮苷提取物浓度升高而降低,当枳实总黄酮苷提取物浓度达到6 000μg/ml及10 000μg/ml时具有统计学意义(P<0.05)。[结论]枳实总黄酮苷提取物对功能性消化不良大鼠近端胃适应性具有一定的调节作用,其作用途径可能与降低近端胃体平滑肌张力有关。[Objective]The aim of this study is to explore the effect of aurantii fructus immaturus flavonoid(AFIF)on the proximal gastric accommodation of functional dyspepsia(FD)rats and preliminary discuss the possible mechanism.[Methods]86ten-day-old male rats were randomly divided into two groups.The model rats(n=62)were given iodoacetamide(IA)through gavage and received tailsqueezed,while the controls(n=24)were only given normal saline.Model group was randomly dividedinto AFIF high-dose group,AFIF middle-does group,AFIF low-dose group and natural recovery group when the rats were eight-week-old,and the four secondary groups received AFIF interventions in highdose,middle-dose,low-dose and normal saline intervention for 14 days respectively.Then gastric accommodation responses to graded gastric balloon distention(GD;1-10 mm Hg)were tested in each group.And the effect of AFIF on resting tension of isolated gastric smooth muscle was tested.[Results]During the ramp phase and the tonic phase,the gastric accommodation of natural recovery group was significantly decreased compared with control group(P〈0.05),while the gastric accommodation of AFIF high-dose group was significantly improved compared with natural recovery group(P〈0.05).And the gastric accommodation of AFIF middle-dose group was significantly improved during the tonic phase(P〈0.05).There was no significant difference founded when comparing the gastric accommodation between AFIF low-dose group and natural recovery group.AFIF concentration-dependent reduced the resting tension of isolated gastric smooth muscle,and statistical significant differences were founded when the dosage of AFIF reached 6 000μg/ml and 10000ug/ml(P〈0.05).[Conclusion]AFIF has a certain regulation effect on proximal gastric accommodation in FD rats,which may be related to the decrease of the resting tension of the proximal stomach muscle.
