摘 要:目的制备抗心绞痛药盐酸曲美他嗪单层渗透泵控释片,考察影响该制剂药物释放的因素以及体内药物代谢动力学。方法通过改变制剂处方的某一因素,根据不同时间药物释放度来考察药物释放情况。结果探索出该药物制剂的优化处方组成,提高了药物释放度,并进行了释药行为的体内外相关性研究。结论自制单层渗透泵片具有良好的体内外相关性,能平稳释放药物,提高了生物利用度,简化了生产工艺。Objective To prepare singer-layer osmotic pump tablets to achieve zero order release pattern, and to investigate the influence factors of drug release and in vivo-vitro correlations. Methods We prepared trimetazidine hydrochloride single-layer osmotic pump tablets, conducted the single factor experiment to ensure the factors which had significant influence on drug release, and recorded the drug release rate of different time to reflect the dissolution. Results The optimum formulation was found to have improved the degree of drug release. Conclusion The singer-layer osmotic pump tablets we prepared have a good in vivo-vitro correlation and sustained drug realease. The bioavailability is improved and the production process simplified.
