摘 要:目的研究安喘舒丸中腺苷和黄芪甲苷在人血浆中的蛋白结合率。方法采用LC-MS/MS法测定腺苷和黄芪甲苷与人血浆蛋白结合率,LC-MS/MS法测定透析内、外液中2种成分质量浓度,并计算血浆蛋白结合率。结果腺苷和黄芪甲苷的线性关系、精密度与准确度、提取回收率和稳定性均符合方法学要求。安喘舒丸低、中、高(30、60、120 mg·L-1)3个浓度中,腺苷的人血浆蛋白结合率分别为(34.78±1.38)%、(35.43±1.04)%、(35.29±1.63)%;黄芪甲苷为(86.69±1.72)%、(86.14±2.37)%、(85.42±2.55)%。结论安喘舒丸中腺苷与人血浆具有低强度蛋白结合率,黄芪甲苷与人血浆具有高强度蛋白结合率。Objective To determine the protein binding rate of adenosine and astragaloside IV in Anchuanshu pills in human plasma. Methods LC-MS/MS method was used to determine the binding rates in vitro, and the determination of adenosine and astragaloside IV both in inner and outer dialysis. Results The linearity, preci- sion and accuracy, extraction recovery and stability of adenosine and astragaloside IV all met the methodology requirements. Within the low, middle and high concentration (30, 60, and 120 mg·L - 1) of Anchuanshu pills, the protein binding rate of adenosine was (34.78±1.38)%, (35.43±1.04)% and (35.29±1.63)%; while the pro- tein binding rate of astragaloside IV was (86.69±1.72)%, (86.14±2.37)% and (85.42±2.55)%. Conclusion In Anchuanshu pill, adenosine has low protein binding rate in human plasma, while astragaloside IV has high binding rate.
